中西医结合治疗对免疫低下肺炎小鼠CD4+CD25+调节性T细胞的影响

             

摘要

目的 建立免疫功能低下小鼠肺炎克雷伯菌肺炎的模型,观察中西医结合治疗对于此种肺炎的CD4+CD25+调节性T细胞表达的影响.方法 采用腹腔注射环磷酰胺的方法建立免疫功能低下的小鼠模型,通过鼻腔滴种肺炎克雷伯菌液制作肺炎模型,用流氏细胞学技术检测各组小鼠给药前、给药后12 h,给药后24 h,给药后36 h小鼠外周血CD4+ T细胞表面Foxp3阳性表达率和CD4+ Foxp3+ T细胞CD25表达强度.结果 各组各时间点的小鼠外周血CD4+ Foxp3+ T细胞CD25表达强度较正常小鼠显著升高(P<0.05),CD4+ T细胞表面Foxp3阳性表达率较正常小鼠明显降低(P<0.05).给药后12 h,中西医治疗组小鼠外周血的CD4+ Foxp3+ T细胞CD25表达强度较模型组显著下降(P<0.05),CD4+ T细胞表面Foxp3阳性表达率较对照组、单纯西药组及单纯中药组显著升高(P<0.05).结论 ①应用环磷酰胺后,小鼠外周血CD4+ Foxp3+ T细胞CD25表达强度显著升高,CD4+ T细胞表面Foxp3阳性表达率明显降低.②中西医结合治疗在免疫功能抑制小鼠肺炎感染模型中可能具有"双向调节免疫"功能的作用,即一方面维持CD4+ T细胞Foxp3阳性表达率,提高机体抗感染能力,同时维持了机体免疫耐受的状态.③中西药结合治疗有改善免疫功能低下感染生存率的趋势.%Objective The aim of the study is to establish an immunosuppressive mouse pneumonia model for Klehsiella pneumonia infection to simulate the status of the recipients who suffer pneumonia. And we use the model to observe the effect on CD4 + CD25 + T cells of which treated hy integrative Traditional Chinese and Western Medicine. Methods The immunosuppressive mouse model is established through intraperitoneal injection of cyclophosphamide. The mouse pneumonia model is established through instill intranasally with suspension of Klebsiella pneu moniae. Flow cytometry was used to detect the ratio of Foxp3 +/CD4 + and the expression of CD25 in CD4 + Foxp3 + T cells of peripheral blood of mice in different therapy groups before and 12.24 and 36 hours after treatment. Results In our study, the ratio of Foxp3 + /CD4 + was significantly lower and the expression of CD25 in CD4 + Foxp3 + T cells was significantly higher than normal mice. In the comhined therapy group, the expression of CD25 in CD4 + Foxp3 + T cells was significantly lower than model group, and the ratio of Foxp3 +/CD4 + was significantly higher than not only the simple western medicine group and the simple Chinese medicine group but also the immunosuppressive mice without pneumonia at the time that was 12 hours after treatment. Conclusions After injected cyclophosphamide the ratio of Foxp3 + /CD4 + was significantly lower and the expression of CD25 in CD4 + Foxp3 + T cells of mice was significantly higher than normal mice. The comhined traditional and western medicine therapy may have two - ways regulation in immune system in this kind of pneumonia model. It can maintain the ratio of Foxp3 + /CD4 + to keep the immune tolerance. At the same time, it can drift the ratio of Th1/Th2 to Th1 type to enhance the immune function.
