辣椒素受体TRPV1在大鼠内脏高敏感性发病机制中的作用

     

摘要

Objective To investigate the role of the the vanilloid receptor TRPV1 in rat model of visceral hypersensitivity. Methods The visceral hypersensitivity model was made by receiving an intracolonic infusion of 0. 5% acetic acid in SD rats from day - 8 after birth(NA group), and the control group was injected with normal saline. TRPV1 inhibitors were injected intraperitoneally 30 minutes after acetic acid sensitization (antagonist group). TRPV1 expression in colon and dorsal root ganglia was observed in two groups of adult rats by immunohistochemical method. Results Neonatal acetic acid treatment resulted in persistent visceral hypersensitivity in adult rats. TRPV1 expression was increased in colon and dorsal root ganglia in NA group compared with control group and antagonist group( P < 0. 05). Conclusion The vanilloid receptor TRPV1 is in-volved in the formation of visceral hypersensitivity induced by colon irritation during neonatal period. The future is expected to be an effective target for the treatment of IBS.%目的:观察辣椒素受体 TRPV1在大鼠内脏高敏感性发生中的作用。方法新生 SD 大鼠出生后第8天起连续2周直肠内给予0.5%醋酸刺激(幼年醋酸刺激组),制作内脏高敏感性模型,对照组则注射生理盐水,另一组在给予醋酸刺激半小时后腹腔内注射 TRPV1拮抗剂(拮抗剂组)。在大鼠成年后,通过免疫组化法观察 TRPV1在两组大鼠结肠、背根神经节表达情况。结果幼年大鼠在给予直肠醋酸刺激后可在成年后发展成持久的内脏高敏感性,其结肠和脊髓背根神经节中 TRPV1表达较对照组明显增多( P <0.05),而 TRPV1拮抗剂组则表达明显下降。结论辣椒素受体 TRPV1参与了发育期幼鼠肠激惹后内脏高敏感性的形成,未来有望成为肠易激综合征治疗的有效靶点。
