混旋聚乳酸/纳米羟基磷灰石板对成骨细胞标志性蛋白mRNA表达及细胞培养环境的影响

         

摘要

Objective To analysis effect of PDLLA/ nano - Ha on the expression of osteoblast marker protein mRNA and cell culture envi-ronment and to provide theoretical basis. Methods The PDLLA/ nano - Ha was prepared into a certain size disinfectant. The fourth generation of fetal rat calvarial cells were inoculated into each material in the culture medium which was changed every other day. The samples were collected at 4,7,14 and 21 days after the change of liquid. Total - 70 RNA was extracted. According to the GenBank rat beta mRNA ALP,mRNA Col -,- actin I mRNA,Cbf alpha 1 mRNA and mRNA BGP full sequence,the use of Manager Clone design primers,each sample set 3 repeat. The concentration of Ca and P were detected and calculated. Results In 4,7,14,21 days,the expression level of Cbf alpha 1 PDLLA/ nano - Ha was significantly higher than that in the control group,and there was significant difference. In 7,14,21 days,the expression level of Col - I mR-NA in PDLLA/ nano - Ha was significantly higher than that in the control group,and there was significant difference (P 0. 05). The 4th days slowly raised,the 14 day again greatly reduced,in the 16 day minimum,PDLLA/ nano - Ha group decreased significantly higher than that in the control group (P 0.05),第4天时缓缓升高,14天时再次大幅降低,在第16天时最低,PDLLA/nano-Ha组下降幅度明显高于对照组(P<0.05),第18天后两组磷酸盐含量均缓缓上升.结论 增殖后期与分化早期PDLLA/nano-Ha能不断促进成骨细胞分化,而在分化晚期(矿化期)却降低了细胞分化的速度.同时PDLLA/nano-Ha还通过改变培养液钙磷浓度促进成骨细胞分泌的体外基质矿化.
