前列腺癌组织内PTTG1基因表达量的变化及其与增殖、侵袭基因的相关性研究

         

摘要

目的 研究前列腺癌组织内垂体肿瘤转化基因l(PTTG1)基因表达量的变化及其与增殖、侵袭基因的相关性.方法 回顾性选择2014年6月至2017年3月期间接受手术切除治疗的前列腺癌患者,收集前列腺癌组织及癌旁组织后抽提RNA.计算前列腺癌组织内PTTG1基因表达量的中位数,按照中位数分为PTTG1高表达及低表达的前列腺癌组织.采用荧光定量PCR试剂盒测定组织内PTTG1基因、细胞增殖基因[增殖性细胞核抗原(PCNA)、细胞周期蛋白(CyclinD1)、Survivin、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)]、细胞侵袭基因[神经型钙黏附素(N-cadherin)、波形蛋白(Vimentin)、上皮型钙黏附素(E-cadherin)、基质金属蛋白酶2(MMP)2、基质金属蛋白酶9(MMP9)]的mRNA表达量.结果 前列腺癌组织内 PTTG1、PCNA、CyclinDl、Survivin、N-cadherin、Vimentin、MMP2、MMP9基因的mRNA表达量显著高于癌旁组织,Caspase-3、E-cadherin的mRNA表达量低于癌旁组织;PTTG1高表达量的前列腺癌组织内 PCNA、CyclinDl、Survivin、N-cadherin、Vimentin、MMP2、MMP9的mRNA表达量均高于 PTTG1低表达量的前列腺癌组织,Caspase-3、E-cadherin的mRNA表达量低于PTTG1低表达量的前列腺癌组织.结论 前列腺癌组织内PTTG1基因呈高表达趋势且与增殖、侵袭基因表达的变化存在密切关系.%Objective To study the changes of the expression of pituitary tumor transforming gene 1 (PTTG1) gene in the prostate cancer and its correlation with the proliferation and invasive genes. Methods Prostate cancer patients received surgery resection in our hospital during June 2014 to March 2017 were selected, RNA were isolated after prostate cancer lesion and cancerous lesions were collected. The median of PTTG1 gene expression in prostate cancer tissues was calculated and divided into PTTG1 high and low expression prostate cancer tissues according to the median. Then mRNA expression of PTTG1 gene, cell proliferation gene, cell invasion gene were determined by fluorescence quantitative PCR assay kit. Results mRNA expression of PTTG1, PCNA, CyclinDl, Survivin,N- cadherin, Vimentin, MMP2, MMP9 in prostate cancer lesion were higher than those in cancerous lesions, mRNA expression of Caspase- 3 and E- cadherin were lower than those in cancerous lesions; mRNA expression of PCNA, CyclinDl, Survivin, N- cadherin, Vimentin, MMP2, MMP9 in PTTG1 high expression prostate cancer were higher than those in PTTG1 lower expression prostate cancer, mRNA expression of Caspase- 3 and E- cadherin were lower than those in PTTG1 lower expression prostate cancer. Conclusion The high expression of PTTG1 gene in prostate cancer lesions is closely related to the changes of proliferation and invasion gene expression.
