针刀疗法对腰椎间盘突出症大鼠背根神经节局部微循环影响的实验研究

         

摘要

Objective:To observe the effect of acupotomy on the changes of blood flow of dorsal root ganglion in L5 and TXB2 and 6-K-PGF1αin plasma of rats with radicular pain of lumbar disc herniation ,so as to explore possible mechanism of acupotomy .Methods:32 male SD rats were randomly divided into a blank group ,a model group ,an acupotomy group and an electroacupuncture group ,with 8 rats in each group .On the eighth day after modeling ,laser doppler flowmeter was taken to record the dynamic graph of blood flow of the dorsal root gangli-on( DRG) in L5 , and the plasma was taken to observe the changes of TXB 2 and 6-K-PGF1αby ELISA.Re-sults:The comparison of the blood flow of dorsal root ganglion among each group was that the model group (46. 56 ±10 .64 ) decreased significantly ( P<0 .01 ) compared with the blank group ( 77 .13 ±20.56 ) ;acupotomy group (70.56 ±19.84) decreased compared with the model group (46.56 ±10.64)(P<0.05) .The compari-son of the ratio of TXB2 and 6-K-PGF1αamong each group was that the model group (1.89 ±0.30)in plasma increased significantly (P<0.01) compared with the blank group (0.77 ±0.19);acupotomy group(1.07 ±0. 18)decreased compared with the model group (1.89 ±0.30) (P<0.01).Conclusion:Acupotomy therapy can improve the local microcirculation of dorsal root ganglion in rats with lumbar disc herniation ,and it may be by regulating the balance of TXB 2 and 6-K-PGF1αto achieve.%目的:观察针刀疗法对腰椎间盘突出症( LDH)大鼠L5背根神经节血流状态及血浆中血栓素B2( TXB2)、6-酮-前列腺素F1α(6-K-PGF1α)含量的影响,探讨针刀疗法对LDH大鼠背根神经节局部微循环影响的可能作用机制。方法:将32只雄性SD大鼠随机分为空白组、模型组、电针组和针刀组,每组8只。造模后第8天用激光多普勒血流仪记录L5背根神经节动态血流量的变化曲线图,并取各组大鼠的血浆,采用双抗夹心ELISA方法检测血栓素B2、6-酮-前列腺素F1α的含量。结果:各组大鼠L5背根神经节局部血流变化的比较,模型组(46.56±10.64)较空白组(77.13±20.56)显著降低,差异有统计学意义(P<0.01),针刀组(70.56±19.84)较模型组(46.56±10.64)显著升高,差异有统计学意义(P<0.05);各组大鼠血浆中TXB2/6-K-PGF1α比值的比较,模型组(1.89±0.30)较空白组(0.77±0.19)显著升高,差异有统计学意义(P<0.01),针刀组(1.07±0.18)较模型组(1.89±0.30)显著降低,差异有统计学意义( P<0.01)。结论:针刀疗法可改善LDH大鼠背根神经节局部微循环,它可能通过调节血栓素B2、6-酮-前列腺素F1α的含量而发挥作用。
