厌氧活性污泥中混合产氢菌群的构建

     

摘要

为了富集产氢菌、去除耗氢菌,用热、酸、碱、三氯甲烷4种方法处理厌氧活性污泥,构建了高效的混合产氢菌群.结果表明,三氯甲烷处理去除耗氢菌的同时不可逆地杀灭了大量产氢菌;而不同程度的热处理、酸处理和碱处理可有效调整菌群结构,富集产氢菌群.经121℃、8 min热处理、pH=4酸处理、pH=10碱处理后,厌氧活性污泥中混合产氢菌群的氢气产率较对照组分别提高了210%、137% 和125%.通过16SrRNA序列分析,表明热、酸或碱这3种简单的富集方法构建了以肠杆菌属为优势菌种的混合产氢菌群,大大提高了氢气产率,同时也为下一步优化菌群打下了良好的基础.%In order to enrich the hydrogen production bacteria and remove the hydrogen consuming bacteria,four pretreating processes (pretreating by heat-shock, acid, base and chloroform, separately) were conducted in batch tests to evaluate and compare their suitability and efficiency for the construction of the mixed hydrogen production flora from digested sludge. The experiments show that the chloroform pretreatment process irreversibly kills lots of hydrogen production bacteria, while the heat-shock, acid and base pretreatments can efficiently adjust the flora construct and enrich the hydrogen production bacteria For fermentative hydrogen production from glucose, compared to the control test, the hydrogen yields increase 210%, 137% and 125%,respectively, by using the digested sludge pretreated with heat-shock (121'C, 8 min), acid (pH=4) and base(pH=I0), separately. The result of 16SrRNA gene sequence analysis indicates that the Enterobacter is the preponderant bacteria in the mixed hydrogen production flora constructed after the enrichment processes with heat-shock, acid or base pretreatment, and it lays the foundation for further optimizing the mixed flora.
