胶原改性聚乳酸的制备及细胞相容性研究

     

摘要

采用将聚乳酸末端羧基酰氯化再与水解胶原活性基团进行反应的方法,利用水解胶原对聚乳酸(PLA)进行了改性,得到胶原改性聚乳酸(CPLA),并采用红外、FITC标记技术、XPS等方法对其结构进行了表征.结果表明:采用上述的方法可以将水解胶原引入聚乳酸分子中.研究了CPLA的亲水性和细胞相容性,结果表明,CPLA的吸水率大于PLA,而水接触角小于PLA,表明CPLA的亲水性明显高于PLA.在细胞外观形态和细胞增殖方面,CPLA与小鼠胚胎成纤维细胞株(3T3)的相容性明显好于PLA.%In this study, collagen-modified PLA (CPLA) was synthesized by the reaction of collagen peptide with acylchlorided PLA which was prepared by acylchloriding the carboxylic groups at the open end of poly (D,L-lactic acid) (PLA), and the characterization of the prepared CPLA was studied by FT-IR, FITC fluorescence labeling method and XPS. The results show that the collagen peptide can be incorporated into the side chain of PLA by means of the said synthesis method. The hydrophilicity and cell compatibility of CPLA were also studied in this paper. Results show that the static water contact angle of CPLA is smaller than that of PLA, while its water absorption ratio is higher than that of PLA, which shows that the hydrophilicity of CPLA is better than PLA. The cell compatibility of CPLA with mouse embryonic fibroblasts (3T3) is significantly better than that of PLA in the respects of cell morphology and cell proliferation.
