PCDH 10基因启动子区高甲基化在结肠癌阶段性发生及进展中的作用

         

摘要

Objective :To explore the role of methylation abnormality of PCDH 10 gene promoter in the staged occurrence and develop‐ment of colon cancer by detecting the methylation status of PCDH 10 gene promoter CpG island from the tissues of colon cancer .Methods:The methylation -specific polymerase chain reaction (MSP) was applied to test methylation status of the PCDH 10 gene promoter CpG island of the colon tissues and the adjacent colon tissues in 56 cases of colon cancer ,and of the normal colon tissues in 15 cases ,analyzing its association with clinical features of colon cancer .Results :The tissues and adjacent colon tissues of the colon cancer and normal colon tis‐sues were examined and quantified .The rates of PCDH 10 gene promoter CpG island methylation abnormality were 57 .1% (32/56) ,10 . 7% (6/56) and 0% among the three groups ,respectively .The differences were statistically significant (P<0 .05) .The methylation sta‐tus of PCDH 10 gene promoter CpG island was significantly correlated to the clinical stages ,the differentiated degree ,and lymph node metastasis of colon cancer (P < 0 .05) .Conclusion:PCDH 10 aberrant gene promoter methylation ,indicated as one of the molecular mechanisms which differentiates normal colon tissues from colon cancer tissues ,may play a key role in the staged occurrence and develop‐ment of colon cancer .PCDH 10 gene promoter CpG island methylation may have the potential as a new tumor diagnosis marker for colon cancer .%目的:本研究通过检测结肠癌组织中PCDH 10基因启动子区CpG岛甲基化状态,探讨 PCDH 10基因启动子区异常甲基化在结肠癌的阶段性发生及进展中的作用。方法:采用甲基化特异性聚合酶链式反应(methylation -specific polymerase chain reaction , MSPCR)检测56例结肠癌及癌旁结肠组织、15例正常结肠组织中PCDH 10基因启动子区CpG 岛甲基化的状态,同时分析其与结肠癌临床特征之间的联系。结果:结肠癌、癌旁结肠组织及正常结肠组织中 PCDH 10基因启动子区 CpG 岛异常甲基化率分别为57.1%(32/56)、10.7%(6/56)和0%,三组之间比较,差异具有统计学意义( P <0.05)。 PCDH 10基因启动子区CpG岛甲基化状态与结肠癌的临床分期、结肠癌的分化程度以及淋巴结转移显著相关( P <0.05)。结论:PCDH 10基因启动子区异常甲基化是结肠癌组织区别于正常结肠组织的分子机制之一,可能在结肠癌的阶段性发生、发展中起着关键的作用。 PCDH 10基因启动子区CpG 岛甲基化可能成为一种新的结肠癌肿瘤标志物。
