复方苦参注射液对人肺腺癌A549细胞株增殖、乙酰肝素酶表达及侵袭能力的影响

摘要

Objective To investigate the possible effects and mechanisms about compound radix sophora flavescentis injection on proliferation and the expression of heparanase(Hpa)of human lung adenocarcinoma cell line A549 in vitro.Method Using the WST-1 to detect the effect on cell growth inhibition rates of all groups after administration of compound radix sophora flavescentis injection;The expressions of Hpa were dectected by immunocytochemical SP assay in each group after 48h period;The invasion a-bility of cells is detected by Matrigel after 48 h period.Results ①The proliferation of A549 cells could not be inhibited by Com-pound radix sophora flavescentis injection for 24 h,however,it could be inhibited by compound radix sophora flavescentis injection for 48 and 72 h.And the energy of inhibition had relationship on concentration of compound radix sophora flavescentis injection with 48 h.Compared with 72 h group,the effect on proliferation inhibition was better in 48 h group. ②Compound radix sophora flavescentis injection could restrain the expression of Hpa of lung adenocarcinoma cell line A549 with the concentration of 0.1~0.5 mg/ml for 48 h. ③Compound radix sophora flavescentis injection could restrain the migration of lung adenocarcinoma cell line A549 with the concentration in 0.2~0.5 mg/ml for 48 h.Conclusion This research shows Compound radix sophora flaves-centis injection has the capacity to inhibits the growth of A549 cell and the effect has the relationship on restraining the expression of Hpa and the migration.%目的：通过体外研究探讨复方苦参注射液对人肺腺癌A549细胞株增殖、乙酰肝素酶（heparanase，Hpa）表达的影响及机制。方法：WST－1法检测不同浓度复方苦参注射液对A549的增殖抑制作用；S－P法测定各组干预48 h后细胞内Hpa的表达；Matrigel体外侵袭试验检测各组细胞侵袭能力。结果：①0.1～0.5 mg／ml浓度的复方苦参注射液作用人肺腺癌A549细胞达48 h开始对细胞增殖有抑制作用，且呈浓度、时间依赖性；相同浓度时干预48 h对细胞的增殖抑制作用优于72 h。②复方苦参注射液作用肺腺癌A549细胞48 h后，药物在0.1～0.5 mg／ml浓度区间内对A549细胞核内Hpa表达有抑制作用。③复方苦参注射液作用肺腺癌A549细胞48 h后，0.2～0.5 mg／ml浓度区间内对A549细胞侵袭基底膜均有抑制作用。结论：本研究结果显示复方苦参注射液能通过抑制肺腺癌A549细胞表达Hpa、抑制A549细胞侵袭基底膜而发挥抗肺腺癌作用。
