消癥活络方对肝纤维化大鼠肝组织α-SMA和TGF-β1表达的影响

         

摘要

目的 观察消癥活络方对肝纤维化大鼠α平滑肌肌动蛋白(α-SMA)和转化生长因子β1(TGF-β1)表达的影响.方法 采用CCl4诱导大鼠肝纤维化模型,将大鼠随机分为6组.通过肝脏病理学HE染色和Masson染色,判断肝脏纤维化程度以及肝组织结构改变;分别采用免疫组化和Western Blot检测肝组织中α-SMA和TGF-β1蛋白的表达.结果 消癥活络方中、高剂量组与秋水仙碱阳性对照组均能明显改善模型大鼠肝组织纤维化程度;抑制大鼠肝脏中α-SMA和TGF-β1蛋白的表达.结论 消癥活络方能够明显改善大鼠肝纤维化程度,其作用机制可能是通过降低TGF-β1的表达,从而抑制肝星状细胞的活化,降低ECM的合成.%Objective To investigate the expression of Xiaozheng Huoluo prescription on α-SMA and TGF-β1 on rats with hepatic fibrosis.Methods Rat liver fibrosis model was induced by CC14.The rats were randomly divided into six groups.To judge the degree of liver fibrosis and the change of liver tissue structure by HE staining and Masson staining;To detect the expression of α-SMA and TGF-β1 protein in liver tissues by using immunohistochemistry and Western Blot method.Results Xiaozheng Huoluo Prescription high dose group,medium dose group and colchicine positive control group can obviously improve the liver fibrosis degree in model rats;The expression of α-SMA and TGF-β1 protein in rat liver was inhibited.Conclusion Xiaozheng Huoluo Prescription can significantly improve the degree of liver fibrosis in rats,Its mechanism may be to reduce the expression of TGF-β1,thereby inhibiting the activation of hepatic stellate cells,reducing the synthesis of ECM.
