食管鳞癌组织中组蛋白去乙酰化酶2的表达及临床意义

         

摘要

目的 探讨组蛋白去乙酰化酶2 (HDAC2)在食管鳞癌组织中的表达及其临床意义.方法 应用实时荧光定量PCR及Western blot法检测30例配对人食管鳞癌(A组)和癌旁组织(C组)中HDAC2蛋白及其mRNA的表达.免疫组化方法检测90例(B组)食管鳞癌组织中HDAC2的表达水平,分析其与临床和病理特性的相关性.结果 A组HDAC2蛋白及其mRNA表达量均显著高于C组(P<0.01).免疫组化结果显示,B组HDAC2蛋白表达阳性率为79%(71/90),HDAC2表达强度与TNM分期和淋巴结转移相关(P<0.05),而与患者年龄、性别、肿瘤大小、肿瘤位置、病理学大体分型、浸润深度及分化程度无关(P>0.05).结论 HDAC2高表达可能是食管鳞癌的特征之一,在一定程度上可以反映肿瘤的恶性程度,可作为判断食管鳞癌患者预后的指标.%Objective To investigate the expressions and significance of histone deacetylase 2 (HDAC2) in esophageal squamous cell carcinoma (ESCC). Methods The expressions of HDAC2 in 30 ESCC tissues (group A) and tumor-adjacent tissues (group C) were detected by RT-PCR and Western blot. The expressions of HDAC2 in 90 ESCC tissues (group B) were detected by immunohistochemistry and the association of HDAC2 and clinicopathological features of ESCC were analyzed. Results The HDAC2 protein and HDAC2 mRNA were significantly higher in group A than those in group C(P<0. 01). The positive expression rate of HDAC2 protein was 79% (71/90) in group B. The HDAC2 expression was positively correlated with TNM stage and lymph node metastasis (P<0. 05), but not associated with gender, age, tumor size and location, pathologic classification,invasion degree and differentiation(P>0. 05). Conclusion High expression of HDAC2 in ESCC may be one of the charactoristics of ESCC, which reflects to a certain extent the malignancy and may be taken as an indicator in evaluating the prognosis of ESCC patients.
