花梨木精油通过嗅觉通路改善小鼠抑郁样行为及其神经递质

         

摘要

目的 探讨花梨木精油改善小鼠抑郁样行为的可行性及其可能机制.方法 将36只C57 BL/6雄性小鼠,采用慢性不可预见性中等强度刺激(CUMS)结合孤养建立抑郁样小鼠模型,将筛选后的模型小鼠随机均分为CUMS+花梨木吸嗅组(花梨木组)、CUMS+盐酸氟西汀组(百忧解组)和CUMS组;另选12只小鼠为对照组.用神经行为学和免疫组化方法检测造模前后小鼠行为学及脑内多巴胺(DA)的变化.结果 与CUMS组比较,花梨木组和百忧解组小鼠在吸嗅治疗2、3和4周时的悬尾安静和强迫游泳时间均明显缩短,而糖水消耗量明显增加(P<0.05).花梨木组和百忧解组在海马、下丘脑、杏仁核DA等阳性反应产物较CUMS组多.结论 花梨木精油吸嗅能够缓解抑郁症小鼠的抑郁样行为,其机制可能是通过嗅觉通路来调节脑的相应中枢而起作用.%Objective To evaluate the feasibility and possible mechanism that rosewood essential oil (REO) can improve depression-like behaviors of mice. Methods With chronic unpredictable moderate-intensity stimulation (CUMS) with isolation, the depression-like model was established in 48 male C57 BL/6 mice. The depressed mice were randomly divided into 4 groups of A (the controls),B(CUMS plus REO sniff), C(CUMS plus fluoxetine) and D(CUMS alone). The neurological behavior was recorded and dopamine ( DA) in the brain was detected using immunohistochemical method before and after modeling. Results Compared to group D, on the 2nd, 3rd and 4th week of the treatment,the times of immobility in tail-suspension test and forced swimming test were all significantly shortened and consumption of sweet water was remarkably increased (P<0. 05). The expression of DA in the hippocampus, hupothalamus and amygdala after sniff treatment was higher in groups of B and C than that in group D. Conclusion REO can attenuate the depression-like behaviors in depressed mice models, which may be through the regulation effect of REO on the central nervous system via olfactory pathway.
