凋亡基因Bcl-2/Bax与高脂饮食诱导大鼠胰岛细胞凋亡的关系研究

摘要

Objective To investigate the relation of apoptosis-related genes with apoptosis of rat pancreatic islet cells due to high fat diet. Methods Apoptotie cells were identified by TUNEL assay, mRNA levels of bcl-2 and Bax were evaluated by immunohistochemical staining. Results High level of fat diet increased the proportion of the apoptotic cell. In the process of apoptosis, mRNA level of Bax increased significantly, while bcl-2mRNA expression decreased. Conclusions High fat diet may induce and exacer-bate diabetes through inducing apoptosis of the islet cells. The change in the ratio of bel-2/Bax mRNA expression may play an important role in apoptosis.%目的 观察凋亡基因Bcl/Bax与高脂饮食诱导大鼠的胰岛细胞凋亡的关系.方法 应用TUNEL法检测高脂饮食诱导的大鼠胰岛细胞凋亡率,应用免疫组化方法检测bcl-2、bax的mRNA的表达.结果 高脂饮食使大鼠胰岛细胞凋亡率明显增加.胰岛细胞凋亡过程中,诱导凋亡基因bax的mRNA表达水平明显升高,抵抗凋亡基因bcl-2 mRNA表达水平明显下降.结论 高脂饮食可能通过诱导胰岛细胞凋亡导致或加重糖尿病,其中bcl-2/bax mRNA表达比率变化可能起重要作用.
