miRNA-146a调控血管平滑肌细胞增殖和凋亡的作用及机制

摘要

目的 探究miRNA-146a调控血管平滑肌细胞增殖、凋亡的作用及相关机制.方法 取SDP级健康雄性SD大鼠作为实验对象,取血管平滑肌细胞(VSMC)消化离心后,转染50 nmol/L miRNA-146a反义寡核苷酸、错义链和同等剂量磷酸盐缓冲液(PBS),进行对照比较.结果 实验组细胞在转染后48 h内VSMC的数目和吸光度值均低于其他两组,细胞凋亡率明显高于其他两组,核因子κBp65、PCNA的表达水平低于其他两组,差异均具有统计学意义(P＜0.05).结论 miRNA-146a可促进VSMC的增殖,抑制细胞凋亡的进行,这一作用机制与核因子κBp65、PCNA表达水平增高有关.%Objective To explore the effect of miRNA-146a on the proliferation and apoptosis of vascular smooth muscle cells and its mechanism.Methods Taking the SDP grade healthy male SD rats as the experimental subjects,After the digestion of vascular smooth muscle cells(VSMC),50 nmol/L transfected miRNA-146a antisense oligonucleotides,missense chain and the same dose of phosphate buffer(PBS)were compared.Results The number and absorbance value of VSMC in the experimental group were lower than those in the other two groups after transfection with 48 h,the apoptosis rate was significantly higher than that of the other two groups,the expression level of nuclear factor κBp65 and PCNA was lower than the other two groups,and the difference was statistically significant(P＜0.05).Conclusion miRNA-146a can promote the proliferation of VSMC,inhibiting the apoptosis of the cells,the mechanism of action is related to the increase of nuclear factor κBp65 and PCNA expression.
