Objective: To research the determination of dosmin on human umbilical vein endothelial cells in order to optimize the concentration of the optimal dosing and optimal dosing time. Method: 1) Human umbilicus vein endothelial cells were divided into two groups of control group and six experimental groups which had six different concentrations of dosmin. MTT method assay was used to detect the activeness of cells and got the optimal dosing. 2) Human umbilicus vein endothelial cells were divided into three groups of A(24h) ,B(48h) ,C (72h) and every group set control group. After adding dosmin to center hours, MTT method assay was used to detect the activeness of cells and got the optimal dosing time. Conclusion; The optimal dosing is 20μg/mL,the optimal dosing time is 48h and the IC50 is 254. 8μg/mL.%目的:测定香叶木苷对脐静脉内皮细胞的影响,优化出最佳加药浓度和最优加药时间.方法:1)将细胞分为对照组和实验组(终浓度10μg/mL、20μg/mL、30μg/mL、40μg/mL、50μg/mL、60μg/mL),MTT法检测细胞活性,根据抑制率和抑制率曲线确定了最佳加药浓度.2)将细胞分为A组(24h)、B组(48h)、C组(72h),每组设置对照,加药(20μg/mL)后分别按照各组时间用MTT法检测细胞活性,根据抑制率和抑制率曲线确定了最优加药时间.结论:经实验选择出的最佳加药浓度为20μg/mL,最优加药时间为48h,其IC50为254.8 μg/mL.
