Three kinds of antistaling agents were prepared with different propotion of sucrose, 8-HQ, citric acid and Aluminum Sulfate, and then used to dispose rice leaves of three-leaf period in vitro.And the leaf blade pigment contents were tested on the same day, 2nd,4th,6th,8th, 10th day of the treatment.Results indicated that lower concentration of antistaling agents was good for leaf blade culture in vitro, but it would promote the senility of leaf blade in vitro when the concentration was too high.As the chlorophyll content of different paddy rice variety decreased at the beginning stage of treatments, carotenoid content increased, with Yinsuo, LTH and Mianhui 2009 as good example.This may be because of the protection function of carotenoid to chlorophyll.Different rice variety had different sensitive concentration of antistaling agents, therefore the kind and concentration of antistaling agents should be carefully determined when conducting paddy rice leaf blade culture in vitro.%用由蔗糖、柠檬酸、硫酸铝、8-羟基喹啉按不同比例配制成3种不同的保鲜剂,处理水稻的离体叶片,分别在处理当天、第二、四、六、八、十天测定其叶片色素含量.结果表明,较低浓度的保鲜剂有利于水稻叶片离体培养,但当超过一定浓度后,反而会促进叶片离体后的衰老;不同水稻品种在处理前期随着叶绿素含量的下降,类胡萝卜素含量有上升趋势,品种银梭、丽江新团黑谷和绵恢2009表现最明显.这可能与类胡萝卜素具有保护叶绿素的功能密切相关;不同水稻品种对保鲜剂敏感浓度的剂量不同,在水稻叶片离体培养时,应根据实际情况来最终确定保鲜剂的种类与浓度,才能达到预期的目标.
