在含有不同浓度蔗糖和聚乙二醇-4000(PEG-4000)的培养基上对黄金梨[Pyrus pyrifolia(Burm.F.) Nakai cv.Whangkeumbae]的花粉离体培养和花粉管生长情况进行了研究.结果表明,当蔗糖和PEG-4000的浓度逐渐升高时,花粉萌发率和花粉管长度都逐渐增加,但浓度过高时表现出一定的抑制作用.当蔗糖浓度为100 g/L时,花粉萌发率达41.3%.花粉管长度达到693 μm;当PEG-4000浓度为15 g/L时,花粉萌发率为39.9%,花粉管长度为675 μm.最适合黄金梨花粉离体培养的培养基组成为H3BO4 50 mg/L+MgSO4 10 mg/L+蔗糖100 g/L+PEG-4000 15 g/L.%The effects of different concentrations of sucrose and PEG-4000 on pollen germination and pollen tube growth of Pyrus pyrifolia (Burm.F.) Nakai cv.Whangkeumbae were studied. The results showed that appropriate amount of sucrose and PEG-4000 was conductive to the pollen germination and the pollen tube growth. But when the concentration was too high, it inhibited pollen germination and pollen tube growth. When the sucrose concentration was 100 g/L, the pollen germination rate and pollen tube length reached 41.3% and 693μm respectively. When the PEG-4000 concentration was 15 g/L, the pollen germination rate and pollen tube length reached 39.9% and 675μm respectively. The concentration of 50 mg/L H3BO4,10 mg/L MgSo4, 100 g/L sucrose and 15 g/L PEG-4000 was suitable to pollen germination and pollen tube growth.
