TIRAP基因多态性与河北地区肺结核易感性的相关性研究

摘要

目的 探讨含Tou-白细胞介素1受体C(TIR)功能区的接头蛋白(TIR domain-containing adapter protein,TIRAP)基因的单核苷酸多态性(SNP)与肺结核遗传易感性之间的相关性.方法 利用SNaPshot技术分析法检测河北地区205例正常人(对照组)和214例肺结核患者(均为汉族人,结核组)TIRAP基因的rs595209,rs7932766,rs8177374和rs8177400位点的多态性,并进行基因型频率和等位基因频率的比较,通过统计学方法分析这些基因的多态性与肺结核易感性之间的相关性.结果 选取的214例结核患者和205例正常人年龄、性别差异无统计学意义(P＞0.05),rs595209,rs7932766,rs8177374和rs8177400位点的基因频率分布均符合Hardy-Weinberg平衡定律,结核组的rs595209位点的基因型及等位基因、rs7932766位点的基因型及等位基因、rs8177374位点的基因型及等位基因与对照组比较,差异均无统计学意义(P＞0.05).对四个位点的等位基因和基因型的频率分布进行统计分析,发现在rs8177374位点等位基因A相对于等位基因G患结核的风险增加了10.9％(OR=1.109,95％CI:0.521～2.361,P=0.788).进一步通过分析发现rs595209位点的GT基因型相对于GG基因型患结核的风险增加了31.4％(OR=1.314,95％CI:0.868 ～1.987),rs8177374位点的GA基因型相对于GG基因型患结核的风险增加了11.3％ (OR=1.113,95％ CI:0.516～2.401),但因为带有GT和GA基因型的人数较少,需要进一步通过加大样本量来进行验证.结论 TIRAP基因的rs595209,rs7932766,rs8177374和rs8177400位点多态性与河北地区汉族人的结核易感性没有显著的相关性.%Objective To investigate the correlation between single nucleotide polymorphisms (SNP) in TIR domain-containing adapter protein gene (TIRAP) and the susceptibility to tuberculosis (TB).Methods The gene polymorphism of TIRAP in gene locus of rs595209,rs7932766,rs8177374 and rs8177400 was analyzed by using SnaPshot technique in 205 healthy subjects (control group) and 214 patients with TB (TB group) of Han people in Hebei province,moreover,the genotypic frequency and allele frequency were compared to analyze statistically the correlation between gene polymorphism and tuberculosis susceptibility.Results There were no significant differences in age and gender between two groups (P ＞ 0.05).The distribution of gene frequency in gene locus of rs595209,rs7932766,rs8177374 and rs8177400 conformed to the Hardy-Weinberg equilibrium law by x2 test.There were no significant differences in genotypic frequency and allele frequency in gene locus of rs595209,rs7932766 and rs8177374As between two groups (P ＞ 0.05).The statistical analysis showed that the risk of suffering TB in rs8177374 locus allele A was increased by 10.9％,as compared with that of allele G (OR =1.109,95％ CI:0.521 ～ 2.361,P =0.788).Further analysis showed that the risk of suffering TB in rs595209 locus GT genotype was increased by 31.4％,as compared with that in GG genetype (OR =1.314,95％ CI:0.868 ～1.987),moreover,the risk of suffering TB in rs8177374 locus GA genetype was increased by 11.3％,as compared with that in GG genetype (OR =1.113,95 ％ C I:0.516 ～ 2.401),however,the number of people with GT genetype and GA genetype was less,which should be identified by increasing sample size.Conclusion The gene polymorphism of TIRAP in gene locus of rs595209,rs7932766,rs8177374 and rs8177400 is not closely correlated with tuberculosis susceptibility in Han people of Hebei province.
