pH胁迫对中华绒螯蟹5种非特异性免疫指标的影响

     

摘要

测定了不同pH(4.5,6.0,7.5,9.0,10.5)胁迫1、4、7、10d后中华绒螯蟹(Eriocheir sinensis)血细胞呼吸爆发(RP)、血细胞破碎物上清液(HLS)酚氧化酶(PO)、血清溶菌酶(LSZ)、酸性磷酸酶(ACP)及碱性磷酸酶(ALP)活性的变化.结果表明:在pH胁迫后第1天,pH4.5组的RP活性显著高于对照组(pH 7.8)和碱性胁迫的3个组,pH 6.0组的PO活性显著高于对照组、pH7.5组和pH 9.0组,pH 10.5组的ACP活性显著高于对照组,pH 6.0组的ALP活性显著高于对照组和碱性胁迫的3个组(P<0.05);在pH胁迫后第7天,pH 6.0组的RP活性显著高于pH 10.5组,pH4.5组的PO活性显著低于对照组和pH 10.5组(P<0.05);血清LSZ对pH胁迫较为敏感,在胁迫后第1天,pH 9.0组显著低于对照组,在胁迫后第4、7、10天,pH4.5、6.0、9.0、10.5组均显著低于对照组(P<0.05).由此可见,RP、PO、LSZ、ACP和ALP可作为pH胁迫对中华绒螯蟹免疫力影响的重要指标.%The changes of immune factors were examined including the activities of respiratory burst ( RP) of haemocyte, phenoloxidase (PO) in haemocyte lysate supernatant (HLS) , lysozyme (LSZ) , acidic phosphatase (ACP) and alkaline phosphatase (ALP) in serum of Chinese mitten crab (Eriocheir sinensis) at 1, 4, 7 and 10 days after different pH (4. 5, 6. 0, 7. 5, 9. 0, 10. 5) stress. The results indicated under 1 day pH stress that the activities of RP in the group of pH 4. 5 were significantly higher than that in control group (pH 7.8) and three alkaline pH stress groups, the activities of PO in the group of pH 6. 0 were significantly higher than that in control group and in the groups of pH 7. 5 and pH 9. 0, the activities of ACP in the group of pH 10.5 were significantly higher than that in control group, the activities of ALP in the group of pH 6. 0 were significantly higher than that in the control group ( pH 8. 0) and in three alkaline pH stress groups (P < 0. 05). Under 7 days pH stress the activities of RP in the group of pH 6. 0 were significantly higher than that in the group of pH 10. 5, the activities of PO in the group of pH 4. 5 were significantly lower than that in control group and in the group of pH 10. 5 (P <0. 05). The activities of LSZ in the group of pH 9. 0 under 1 day pH stress and in the group of pH 4.5, 6.0, 9.0 and 10.5 at 4, 7 and 10 days after pH stress were significantly lower than that in control group (P< 0.05). The main immune parameters, such as RP, PO, LSZ, ACP and ALP can be used as early-warning signals in E. Sinensis cultivation.
