Objective To analyze the performance of GlobalFiler™ kit testing bones of corpses. Methods Silicon bead method was used to extract DNA from 50 samples of bones and teeth that were all required to determine their source with 11 of them waiting for the triplet and the rest 39 ones the duo paternity testing. The GlobalFiler™ kit and Identifiler Plus kit were employed to evaluate their testing competence and cumulative paternity index (CPI) values. Results GlobalFiler™ kit successfully detected the STR types of the 50 samples. For identification of cadaveric source of the 11 triplet cases, although both kits showed to have CPI values greater than 10,000, the GlobalFiler™ kit performed even better with its CPI larger than 108 in 10 out of the 11 cases, much higher than that of Identifiler Plus kit. For the 39 duo cases, GlobalFiler™ kit again elevated its CPI ≥ 10,000 while the Identifiler Plus kit was only 30. Furthermore, GlobalFiler™ kit can expose the genotyping of 21 autosome, Amelogenin, Y-indel and DYS391, especially with the last two exclusive loci for the kit to be capable of determining gender when samples are even of Y-allele-absence in Amelogenin. Conclusions GlobalFiler™ kit, capable of amplifying more loci in a single run of amplification than the widely used Identifiler Plus kit, has greatly increased the CPI for identifying unknown bodies. Moreover, its additional Y-chromosome loci can help determine gender more effectively. Therefore, GlobalFiler™ kit is high applicable in identifying unknown bodies.%目的:分析 GlobalFiler™试剂盒在白骨化尸体身源认定中的技术性能指标。方法采用硅珠法提取50份尸源认定中的骨骼、牙齿样本 DNA,其中三联体亲权鉴定11份,二联体亲权鉴定39份,比较 GlobalFiler™试剂盒和 Identifiler Plus 试剂盒检验结果。结果 GlobalFiler™试剂盒成功检出了50份样本 STR 分型。11份三联体的尸源认定中,两种试剂盒计算得到的 CPI 值均能达到10000以上,但 GlobalFiler™试剂盒得到的累积父权指数(CPI 值)更高,其中10份能达到108以上。39份二联体的尸源认定中,GlobalFiler™试剂盒得到的 CPI 均能达到10000以上,而 Identifiler Plus 试剂盒得到的 CPI 值能达到10000以上的只有30例。而且GlobalFiler™试剂盒扩增得到21个常染色体,Amel 基因座和 Y-indel、DYS391的分型信息,其特有的 Y-indel和 DYS391两个基因座,确保在 Amelogenin 基因座 Y 等位基因丢失的样本中,可以成功检出性别基因,防止性别误判。结论 GlobalFiler™试剂盒扩增可以获得比目前常用的 Identifiler Plus 试剂盒更多基因信息,大大提高了身源鉴定中的 CPI 值,而且可以有效避免 Amelogenin 基因座 Y 等位基因丢失时导致的无名尸体性别误判。因此,Globalfiler 试剂盒在白骨化尸体身源认定中具有较高的应用价值。
