固定化嗜麦芽寡养单胞菌的制备及其降解羽毛废弃物的初步研究

         

摘要

The immobilized Stenotrophomonas maltophilia DHHJ cells were produced by adsorption method with self-made chitosan bead as carries. The obtained immobilized cells were used to biodegradation of waste feather. Results showed that the chitosan bead produced with 3. 0% (mass ratio) of chitosan achieved the best morphology. Over by high pressure sterilization, the immobilized cells occurred in porous appearance with high water-absorbing capacity, its elasticity was also improved, that were favorable for the immobilization and growth of microbial. Adding 10 g of chitosan beads into 100 mL of cell suspension for 2 d, the obtained immobilized cells could release the most effective living bacteria of 7. 0 × 106 cfu/mL during the fermentation. During ten cycles or 40 d of fermentation, the biodegradation rate of feather and keratinase enzyme activity in fermented liquid maintained over 80% of maximum value. The immobilized cells could degrade waste feather efficiently and steadily in long time, they had the potential of continuous feather fermentation. The optimum fermentation conditions for feather degradation were chitosan dosage 50 mg/mL, waste feather 25 mg/mL, shaking speed 130 r/min and fermentation 5 d.%以自制壳聚糖小球为载体,采用吸附固定法制备固定化嗜麦芽寡养单胞菌(Stenotrophomonas maltophilia DHHJ),并将其用于羽毛废弃物降解.结果表明,采用3.0%(质量分数)的壳聚糖制成的壳聚糖小球的形态较好,经高压灭菌后弹性增加,呈多孔状,有很强的吸水性,有利于细菌的吸附固定和生长;在细胞稀释液中加入10%(质量分数)的壳聚糖小球,固定时间设为2d时,制得的固定化小球在发酵过程中释放的有效活菌数最多,可达7.0×106 cfu/mL;在连续10次、共计40 d的序批发酵降解过程中,固定化菌的羽毛降解率和发酵液中的角蛋白酶酶活均保持在最大值的80%以上.固定化菌的发酵稳定性较强,能在较长时间内稳定、有效地降解羽毛,具有连续发酵潜力;固定化菌发酵降解羽毛的最佳工艺条件为:小球投加量50 mg/mL,羽毛投加量25mg/mL,摇床转速130 r/min,发酵5d.
