氨基修饰聚醚醚酮表面及其成骨细胞相容性

         

摘要

首先采用硼氢化钠将聚醚醚酮(PEEK)表面的羰基还原为羟基,获得羟基化预处理的PEEK,再用3-氨基丙基三乙氧基硅烷与其进行反应,将氨基(—NH2)引入至PEEK表面以改善其细胞相容性.用X射线光电子能谱和力学性能测试对表面化学处理的PEEK进行表征,同时,用MC3T3-E1成骨细胞评价表面氨基修饰的PEEK的细胞相容性.结果表明,硼氢化钠成功地将PEEK表面的羰基还原为羟基,—NH2成功地引入至PEEK表面;表面化学处理前后PEEK的拉伸及弯曲强度未发生明显变化;和未修饰的PEEK表面相比,—NH2修饰的PEEK表面能显著促进成骨细胞的粘附、铺展及增殖.%Polyetheretherketone (PEEK) surface activation of carbonyl being reduced to hydroxyl was done with sodium boro-hydride. And then treatment of the hydroxylation-pretreated PEEK samples with a substituted 3-aminopropyltriethoxysilane solution endowed PEEK surface with amino (-NH2) to enhance the cytocompatibility of PEEK surface. X-ray photoelectron spectroscopy and mechanical properties tests were employed to characterize the surface chemical modified PEEK samples. Meanwhile,MC3T3-E1 osteoblast was used to evaluate in vitro biocompatibility of amino-modified PEEK surface. The results show that the carbonyl groups on PEEK surface are successfully reduced to hydroxyl groups by sodium borohydride and —NH2 is successfully introduced on PEEK surface. Surface modifications do not affect the tensile strength and flexural strength of PEEK. MC3T3-E1 osteoblast adhesion, spreading and proliferation on amino-modified PEEK surface are remarkably improved compared to intact PEEK.
