Objective To establish D-galactose induced aging mouse model and investigate thymic-associated genes expression. Meth-ads Thymic-associated genes expression were measured by real-time RT-PCR method valuing directly thymic microenvironment. Results Compared with control group, thymic-associated genes expression ratios of D-gal group were LM02: 0.94±0.76, FoxnI: 0.45±0.32, IL-7: 0.69±0.7. Concusions In contrast to control group, thymus microenvironment negative gene expressions in D-Gal group have no ob vi ous change, but some necessary gene expressions decrease. So, D-Gal treatment can induce thymus involvation.%目的 建立D-半乳糖致衰老小鼠模型,分析其胸腺相关基因表达的研究.方法 D-Gal模型组小鼠皮下注射剂量为500 mg/kg·d的D-半乳糖溶液,连续注射8 W;实时定量逆转录PCR方法对D-Gal模型组小鼠的几种胸腺状态和功能相关基因的表达进行相对定量以直接评价胸腺微环境.结果 D-Gal模型组与对照组相比,胸腺相关基因表达变化比值为:LM02:0.94±0.76,Foxnl:0.45±0.32.IL-7:0.69±0.7.结论 D-Gal模型组与对照组相比,胸腺负调控因子的表达无明显变化,而胸腺微环境上皮细胞和胸腺细胞分泌及必需的因子的基因表达显著下降,因而在个体水平上,D-Gal破坏了胸腺微环境的稳定性和功能性,使小鼠胸腺发生明显退化.
