简便快速测定人体血浆中吗啡含量的RP-HPLC法

摘要

Objective To establish determination of morphine in plasma by RP-HPLC.Methods Column was Diamonsil? C18 (250×4.6 mm,5μm);Mobile phase was MeOH : Buffer (80:20,V/V),the flow rate was 1.3 mL/min;UV wavelength was 233 nm.Results Within a concentration range of 1.8-921.6 ng/mL,the standard curve of Morphine was C=0.962 7F-0.019 6 and had a good linearity(r=0.999 7,n = 8).The lower limit of quantitation was 1.8 ng/mL.Conclusion It appears to be a rapid,convenient,sensitive and reproducible method for clinical research and adverse reaction monitor of Morphine.%目的 建立RP-HPLC法测定人体血浆中吗啡的含量.方法 色谱柱为美国Diamonsil(250×4.6 mm,5 μm) C18反相柱;流动相为甲醇-缓冲液(5 mL三乙胺加入1 000 mL水中,磷酸调pH值7.0)=80∶20,流速为1.3 mL/min;检测波长233 nm.结果 吗啡血浆浓度在1.8~921.6 ng/mL范围内,线性关系良好,标准曲线为C=0.962 7 F-0.019 6(r=0.999 7,n=8),定量下限1.8 ng/mL.结论 本方法 具有快速、简便、灵敏度高、重现性好等优点,可以用于吗啡临床研究和不良反应监测.
