NK细胞上清液提高CTL细胞对肿瘤细胞杀伤力及机制的研究

摘要

Objective To investigate whether supernatants from activated NK cells endows T cells on melanoma cells function. Methods Natural killer(NK) cells were cultured and activated with IL-12,IL-18 ,and both with IL-2,3 days later take out superna-nts,then culture with M05 tumor cells overnight,then check MHC class I (H-2Kb) expression,and as targets for phytohemagglu-tinin and IL-2 activated killer T (CTL) cells for cytotoxic assay. Results These supernatants increase M05 cells MHC class I expression, and CTL cells to M05 cells cytotoxicity. Conclusion Supernatants from NK cells will induce increased MHC expression on target cells and will enhance CTL functionality.%目的 研究从白细胞介素2(IL-2)、IL-2+IL-12、IL-2+IL-18、IL-2+IL-12+IL-18活化的NK细胞提取的上清液是否能提高细胞毒性T细胞(CTL)对黑色素瘤细胞的杀伤率及其机制.方法 提取NK细胞,IL-2 6 000 IU/mL、IL-2 6 000 IU/mL + IL-12 10 ng/mL,IL-2 6 000 IU/mL+IL-18 100 ng/mL,IL-2 6 000 IU/mL+IL-12 10 ng/mL+IL-18 100 ng/mL活化,3 d后提取上清液,与M05细胞培养过夜,流式细胞仪检测肿瘤细胞表面Ⅰ类组织相容性抗原(MHC Ⅰ)表达,以其为靶细胞检测CTL细胞对M05细胞杀伤率.结果 NK细胞上清液提高M05细胞表面MHC Ⅰ分子表达及CTL细胞对M05细胞杀伤率.结论 NK细胞上清液能够增强CTL对肿瘤细胞的杀伤作用.
