Objective To explore the expression level of Candida albicans fluconazole resistance genes CDR1 ,CDR2 and MDR1 in susceptible and resistant strains .Methods Candida albicans was isolated from the detection samples and identified ,115 strains were selected and determined MIC of Candida albicans by the microdilution method .Then PCR was adopted to detect the expression level of Candida albicans resistance genes in susceptible and resistant strains .Results The half inhibitory range in 115 strains of Candida albicans to fluconazole was 5 μg/mL ,90% of antibacterial range was 126μg/mL ;among them ,76 strains were sensitive ,39 strains were resistant ,the sensitivity rate was 66 .1% ,the resistant rate was 36 .8% .The CDR2 gene △ CT was 9 .33 ± 3 .21 for the sensitive strains and 7 .49 ± 2 .54 for the resistant strains ,two groups had statistically significant difference (t=2 .659 ,P<0 .05);the expression levels of CDR1 gene and MDR1 gene had no significant difference between the susceptible strains and the resistant strains(P>0 .05) .Conclusion Drug resistance of Candida albicans is more complicated process ,in which ,its resistance to flucon-azole is related with the high expression of CDR2 and has no relation with the expression of CDR1 and MDR1 .%目的:探讨对氟康唑耐药白念珠菌分离的耐药基因CDR1、CDR2和M DR1在敏感株和耐药株中表达水平。方法从送检样本中分离并鉴定出白念珠菌,选取其中115株采用微量稀释法测定对氟康唑耐药的白念珠菌最小抑菌浓度(M IC )值,并采用PCR方法检测白念珠菌耐药基因在敏感株和耐药株中表达情况。结果115株白念珠菌对氟康唑的半数抑菌范围为5μg/mL ,90%抑菌范围为126μg/mL ;其中76株敏感,39株耐药,敏感率为66.1%,耐药率为36.8%。CDR2基因△CT值敏感株组为9.33±3.21,耐药株组为7.49±2.54,两组比较差异有统计学意义(t=2.659,P<0.05);CDR1基因和MDR1基因的表达水平在敏感株和耐药株中比较差异无统计学意义(P>0.05)。结论白念珠菌耐药是个较为复杂的过程,其中白念珠菌对氟康唑耐药与CDR2高表达有关,与CDR1和MDR1表达无关。
