番茄中一个F-box/FBA基因的表达分析及载体构建

     

摘要

通过生物信息学分析从番茄基因数据库中筛选出一条全长cDNA序列,命名为SlFbf(GenBank登录号:AK326236.1).通过设计特异引物克隆该基因.SlFbf全长1464bp,编码381个氨基酸,N端含有F-box结构域,C端含有FBA_1结构域,在基因组序列中无内含子.半定量及定量RT-PCR分析结果表明,该基因在番茄的根、茎、叶、花蕾、花、青果、黄果及红果中均有表达,且在开花当天的雄蕊中表达最强,推测该基因在雄蕊发育中起着重要作用.同时构建了番茄SlFbf基因的正、反义植物表达载体,为深入研究该基因功能奠定了基础.%A full -length gene named SIFbf was cloned from KatTom full -length cDNA database.and was characterized to be intron-free and have typical F-box and FBA_1 domain. Semi-qPCR analysis showed that SIFbf expressed in all tissues including root, stem, leaf, alabastrum, flower, green fruit, yellow fruit and red fruit, but the highest in flower. Real time RT-PCR results demonstrated that SIFbf transcripts were intense in stamen among the four components of flower at bloom. These results suggested that SIFbf maybe participate in stamen development. For studying the function of SIFbf by trangenic method, the sense and antisense plant expression vector of SIFbf were constructed, named pCAMBIA1301-Fbfs, pCAMBIA1301-Fbfas separately.
