益气养阴活血方对2型糖尿病大鼠胰岛素抵抗及脂肪组织microRNA-29的影响

摘要

Objective:To investigate the effects of Yiqi Yangyin Huoxue Fang on insulin resistant and miR-NA-29 expression in rats with type 2 diabetes mellitus.Methods:Experiment was divided into normal,model,Yiqi Yangyin Huoxue Fang high,middle and low dose groups and positive group (Jinlida),models were copied by feeding with high fat diet together with peritoneal injection of STZ,every medicine group was givern different doses of Yiqi Yangyin Huoxue Fang and Jinlida particle for 4 weeks,then fasting plasma glucose (FBG) and fasting insulin(FINS) were tested,the degree of insulin resistance was measured by Homeostasis model assessment -insulin resistant(HOMA-IR) and insulin sensitivity index(ISI).RT-PCR method was applied to determine the expression of miRNA-29 in rat' s adipose.Results:Compared with the model group,the level of FBG and FINS in different doses of Yiqi Yangyin Huoxue Fang groups were decreased obviously(P ＜0.01),ISI was increased obviously(P ＜ 0.01),HOMA-IR was decreased obviously(P ＜ 0.01),and miRNA-29 expression in adipose was reduced significantly (P ＜ 0.05).Conclusion:Yiqi Yangyin Huoxue Fang can ameliorate the insulin resistance of T2DM rats,and its mechanism is probably related to the effect of decreasing the expression of miR-NA-29.%目的:观察益气养阴活血方对2型糖尿病大鼠胰岛素抵抗及脂肪组织中microRNA-29表达的影响.方法:实验分正常对照组、模型组、益气养阴活血方高中低剂量组和津力达阳性对照组,以高脂饮食加小剂量链脲佐菌素(STZ)诱导形成2型糖尿病胰岛素抵抗大鼠模型,各药物干预组大鼠分别予益气养阴活血方高、中、低剂量和津力达颗粒灌胃4周后,检测空腹血糖(FBG)、血清胰岛素(FINS)含量,并计算胰岛素抵抗指数(HOMA-IR)和胰岛素敏感指数(ISI);采用RT-PCR法检测脂肪组织中microRNA-29的表达.结果:与模型组比较,益气养阴活血方能明显降低模型鼠FBG和FINS水平(P＜0.01),提高ISI (P＜0.01),降低HOMA-IR(P＜0.01),并显著降低大鼠脂肪组织中miR-29表达(P＜0.01).结论:益气养阴活血方对2型糖尿病大鼠胰岛素抵抗状态有改善作用,其改善作用可能是通过降低microR-NA-29的表达实现的.
