长期有氧运动对代谢综合征大鼠炎症及心肌组织过氧化物酶体增殖物激活受体α表达的影响

         

摘要

目的:研究有氧运动对代谢综合征(MS)大鼠炎症反应的影响以及心肌组织过氧化物酶体增殖物激活受体α(PPARa)的表达,探讨运动对MS心血管炎症反应调节的可能机制.方法:4周龄SD雄性大鼠随机分为基础对照组(C)和高脂高盐饲料喂养组.18周后将MS大鼠随机分为模型对照组(MC)、模型运动组(ME).运动组大鼠行60min/d、27m/min、5%坡度跑台训练12周.ELISA法测定血清IL-6、TNF-α、MCP-1含量;荧光定量R-PCR测定心肌组织PPARαmRNA表达、Western Blot测定心肌PPARα蛋白含量.结果:①与C组相比,MC组血清IL-6、TNF-α、MCP-1含量显著升高(P< 0.05、P<0.01、P<0.01)、MC组心肌PPARa mRNA和蛋白表达显著降低(P<0.05);②与MC组相比,ME组大鼠血清IL-6、TNF-α、MCP-1含量显著降低(P< 0.05、P<0.01、P< 0.01)、ME组心肌PPARα mRNA和蛋白表达显著升高(P< 0.05、P<0.01).结论:MS大鼠炎症反应明显,长期有氧运动干预能有效降低其炎症反应,PPARα可能介导了机体的炎症反应.%Objective:To research the effects of aerobic exercise on the cardiovascular inflammatory response in rats suffering from metabolic syndrome (MS).And its possible mechanism will be studied through observing the changes of peroxisome proliferator-activated receptor alpha (PPARα) expressing in the myocardial tissues.Method:4-week old male SD rats were randomly divided into control group (C) and high-fat-high-salt feed group.After 18 weeks,MS rats were randomly divided into model control group(MC) and model exercise group(ME).Rats in ME group were forced to run on a treadmill for 12 weeks with the intensity of 60min/d,27m/min and 5% gradient.Serum IL-6,TNF-α,and MCP-1 were detected by ELISA.The expression of PPARα mRNA in myocardium was determined by RT-PCR.Western Blot was applied to detect the protein expression of PPARα in myocardium.Result:①Compared with C group,the MC group showed significant increase in serum IL-6,TNF-α,MCP-1 (P<0.05,P<0.01,P<0.01) and significant decrease in PPARαt mRNA and protein expression in myocardium (P<0.05).②Compared with MC group,the ME group showed significant decrease in serum IL-6,TNF-α,MCP-1 (P<0.05,P<0.01,P<0.01) and significant increase in PPARα mRNA and protein expression in myocardium(P<0.05,P<0.01).Conclusion:The inflammation in MS rats occurred obviously.Long-term aerobic exercise can effectively improve it.PPARα mediates the inflammatory in MS rats potentially.
