未破裂颅内动脉瘤患者外周血内皮祖细胞数量的研究

             

摘要

目的 研究颅内动脉瘤与外周血中内皮祖细胞(EPCs)数量的关系.方法 选择未破裂颅内动脉瘤患者24例和正常对照者16例,用密度梯度离心法获取外周血单个核细胞,加入培养基进行选择性培养,分析14 d后细胞形成的集落数,并对贴壁细胞进行细胞鉴定、分析和计数.激光共聚焦显微镜下观察其吞噬荆豆凝集素和乙酰化低密度脂蛋白的内皮功能;用流式细胞仪检测CD34、CD133和血管内皮生长因子受体2(VEGFR-2)三抗阳性细胞并计数.结果 颅内动脉瘤患者外周血EPCs计数较对照组明显减少,流式分析结果显示三抗阳性细胞数比例有显著性差异.结论 颅内动脉瘤患者外周血EPCs的数量较无动脉瘤者明显降低.%Objective To investigate the morphology, growth and counts of endothelial progenitor cells (EPCs) in peripheral blood of patients with unruptured intracranial aneurysms (UIAs). Methods Twenty-four patients with UIAs and 16 controls were chosen. Mononuclear cells from peripheral blood were harvested by density gradient' centrifugation. Then the cells were selectively cultured. On 14 d, the colony counts were analyzed and the adherent cells were identified, analyzed and counted. The biological functions of adherent cells were examined by the adsorption of ulex europaeus agglutinin-1 ( UEA-1) labeled by fluorescein isothiocyanate (FFTC) and acetylated low-density lipoprotein (Dil-acLDL) internalization and the positive double dyeing cells indicated the endothelial progenitor cells under differentiation. The endothelial progenitor cells were examined by flow cytometry for CD34, CD133 and vascular endothelial growth factor receptor 2 ( vEGFR-2). Results The count of endothelial progenitor cells in peripheral blood was significantly lower in patients with UIAs than that of controls. CD34 VCD133 VVEGFR-2 * cells were significantly decreased in patients with UIAs compared with controls. Conclusion The count of endothelial progenitor cells in peripheral blood is decreased in patients with intracranial aneurysm compared with non-aneurysm patients.
