DNA依赖蛋白激酶与切除修复交叉互补基因1在上皮性卵巢肿瘤组织中的表达及意义

         

摘要

目的 分析DNA依赖蛋白激酶 (DNA-PKcs)、切除修复交叉互补基因1 (ERCC1)在上皮性卵巢肿瘤组织中的表达及其意义.方法 采用免疫组化SP法检测65例上皮性卵巢癌、20例卵巢交界性肿瘤、18例卵巢良性肿瘤及15例正常卵巢组织中DNA-PKcs、ERCC1的表达,并分析其异常表达与卵巢恶性肿瘤临床病理特征的关系;分析DNA-PKcs与ERCC1表达的相关性.结果 DNA-PKcs在卵巢癌(66.15%)、交界性卵巢肿瘤组织(55.00%)中的阳性表达率高于卵巢良性肿瘤(11.11%)及正常卵巢(6.67%),差异有显著性,ERCC1在卵巢癌(67.69%)、交界性卵巢肿瘤组织(60.00%)中的阳性表达率高于卵巢良性肿瘤(16.67%)及正常卵巢(13.33%),差异有显著性;DNA-PKcs、ERCC1表达与残留灶大小有关,与FIGO分期、病理类型、病理分级及淋巴结转移无关;DNA-PKcs与ERCC1表达呈正相关.结论 DNA-PKcs、ERCC1表达与卵巢癌的发生发展有关,在促进卵巢癌的浸润转移过程中可能起重要作用.%Objective To explore the expression and clinical significance of DNA-PKcs and ERCC1 in epithelial o-varian tumors. Method Immunohistochemistry SP method was used to detect the expression of DNA-PKcs and ER-CC1 in 65 cases of epithelial ovarian cancer, 20 patients with borderline ovarian tumors, 18 patients with benign o-varian tumors and 15 normal ovarian tissue. The relationship between the expression of DNA-PKcs and ERCC1 and some clinicopathologic characteristics in ovarian cancer were analyzed. Meanwhile, the correlation of the DNAPKcs and ERCC1 expression was study as well. Result The positive rate of DNA-PKcs in ovarian cancers (66. 15% )and borderline ovarian tumors (55. 00% ) were higher than in benign ovarian tumors (11. 11% ) and normal ovaries (6. 67% ) , the differences were statistical significance, the positive rates of ERCC1 in ovarian cancers and borderline ovarian tumors (67. 69% , 60.00% ) were higher than in benign ovarian tumors ( 16. 67% ) and normal ovaries ( 13. 33% ) ,the differences were statistical significance;Evidence was found that the expression of DNA-PKcs and ERCC1 was correlated to the diameter of residual tumor, and there was no significant correlation with the age,FIGO stage, pathological type, pathological grade and lymph node metastasis; The expressions between DNA-PKcs and ERCC1 showed positive correlation. Conclusion The expression of DNA-PKcs and ERCC1 was correlated with the occurrence and development of ovarian cancer, and might play an important role in promoting the process of ovarian cancer invasion and metastasis.
