目的 探讨klotho基因转染对脂肪间充质干细胞(ADSCs)增殖的影响.方法 将含有klotho全长基因的真核质粒转染至从脂肪组织分离的ADSCs中,采用CCK-8法测定转染后细胞增殖率,western blot法检测转染后细胞周期蛋白D1的表达变化,流式细胞术检测转染前后ADSCs的细胞周期变化.结果 转染klotho基因96h后ADSCs的细胞增殖率为170.50%;流式细胞术检测结果显示,ADSCs转染klotho基因S期和G2期细胞明显增多;western blot分析显示ADSCs转染klotho基因组细胞周期蛋白D1增高.结论 转染klotho基因有促进ADSCs增殖的作用.%Objective To investigate proliferation of ADSCs induced by kiotho gene transfection. Methods ADSCs were isolated from liposuction specimens and transfected with the kiotho gene expression piasmid. The proliferation rate of ADSCs was detected by CCK-8,thc expression of cyclinD and the cell cycle were detected by western blot and flow cytomctry. Results When ADSCs were transfected with the vector containing klotho gene, the proliferation rate of ADSCs was 170. 50% after 96h of transfection. Flow cytomctric analysis showed that the number of cells in S and G2 phase had increased significantly. Western blot analysis indicated that cyclinDl expression in klotho gene transfection group had also increased. Conclusion klotho gene can promote the proliferation of the ADSCs.
