酪蛋白激酶CK2抑制剂TBB增加硼替佐米诱导的结肠癌细胞系HT29细胞凋亡

     

摘要

目的 探讨酪蛋白激酶CK2抑制剂TBB对硼替佐米诱导的结肠癌细胞凋亡的影响.方法 选用人结肠癌细胞系HT29细胞,MTT法检测单独使用硼替佐米和联合四溴苯三唑(TBB,CK2抑制剂)对HT29细胞增殖抑制作用,流式细胞术检测单独使用硼替佐米和联合TBB对HT29细胞凋亡的影响,Western blot方法检测硼替佐米和联合TBB处理的HT29细胞Cleaved Caspase-3的表达情况.结果 MTT结果显示硼替佐米能抑制HT29细胞增殖并呈现剂量依赖性,硼替佐米与TBB联合使用更能抑制增殖.单独使用硼替佐米能诱导细胞凋亡,两者联合的细胞凋亡效应更加明显.硼替佐米能促使Cleaved Caspase-3表达上调,两者联合使用会进一步增加Cleaved Caspase-3的表达.结论 硼替佐米能诱导HT29细胞凋亡,而CK2促进了硼替佐米诱导的细胞凋亡.%Objective To explore the effect of bortezomib combined with TBB on the growth inhibition and apoptosis of cells in vitro.Methods Colon cancer cell line HT29 cells was used as research object.MTT was employed to measure inhibiting action of bortezomib and/or TBB on HT29 cells.The degree of apoptosis in HT29 cells was detected by flow cytometry.The protein expression levels of Cleaved Caspase-3 was detected using Western blot in HT29 cells treated with bortezomib and/or TBB.Results MTT assay showed that bortezomib alone inhibited the proliferation of HT29 cells in a dose dependent manner and TBB enhanced inhibitory action of bortezomib.Flowcytometry results show that Bortezomib induced HT29 cells apoptosis,where apoptotic proportion of combination group was markedly increased.Bortezomib up regulated the expression of Cleaved Caspase-3 in HT29 cells.Cleaved Caspase-3 was enhanced in HT29 cells treated with bortezomib combined with TBB.Conclusion Bortezomib alone can inhibit proliferation and induce apoptosis of HT29 cell,TBB treatment enhanced depressing effect and apoptosis rate induced by bortezomib.
