Objective:To observe the effect of compound Chinese medicine Tangnaikang on related loci in ERK signal trans-duction pathway in rat mesangial cells induced by high glucose. Methods: MCs of rats were induced by normal glucose, high glucose, Pioglitazone and Tangnaikang decoction at different dosage, estimated mRNA expression of cPLA by RT - PCR and the protein content of p - ERK、PCK - α、c - fos by western blot. Results: Hyperglycemia reinforced the expression of cPLA and the protein content of p -ERK、PCK - α、c -fos, Tangnaikang decoction could countered the effect and showed dependence of dose. Conclusion:Tangnaikang decoction could protect mesangial cells through countering related loci in ERK signal transduction pathway.%目的:观察复方中药糖耐康对高糖诱导大鼠系膜细胞中胞外调节蛋白激酶通路相关蛋白表达的影响.方法:大鼠肾小球系膜细胞按1×105/孔接种24孔板,实验分为6组:正常葡萄糖组、高糖对照组、中药糖耐康高、中、低浓度干预组、西药吡格列酮组.每组设平行孔4孔,培养24 h后检测指标.用RT-PCR法测定各组细胞cPLA mRNA表达,用蛋白质印迹杂交方法(Western Blot)检测p-ERK、PCK-α、c-fos蛋白的表达.结果:治疗组cPLA mRNA、p-ERK、PCK-α、c-fos蛋白表达的水平比模型组均有不同程度的降低(P<0.01,P<0.05).结论:糖耐康对糖尿病肾病肾脏的保护作用,可能是糖耐康抑制了PKC的活化,阻止了DAG-PKC转导途径向通过ras依赖的ERK的传导,并阻止了转录因子c-fos的活化,使不通过ras途径激活ERK的PKC无法激活相关转录因子.
