舒郁宁心汤对抑郁模型大鼠海马区脑源性神经营养因子和酪氨酸激酶B表达的影响

             

摘要

Objective To investigate the effect of Shuyu Ningxin decoction on the expression of BDNF and TrkB in hippocampus of chronic stress depression rats, and explore its antidepressant mechanism Methods Sixty SD rats were selected and randomly divided into control group, model group, fluoxetine group, high dose of Shuyu Ningxin decoction group, middle dose of Shuyu Ningxin decoction group and low dose of Shuyu Ningxin decoction group. Except control group, the other groups were given the chronic unpredicted mild stress for 21 days to induce depression. Then the rats was given corresponding drugs for 21 d. The levels of BDNF and TrkB in hippocampus were investigated with immunohistochemistry technique. Results Compared with control group, the level of BDNF and TrkB in model group was significantly lower (P<0.01). Compared with model group, the level of BDNF and TrkB in fluoxetine group, high dose of Shuyu Ningxin decoction group and middle dose of Shuyu Ningxin decoction group were significantly increased (P<0.05). Conclusion The anti-depression effect of Shuyu Ningxin decoction may be related to its function of improving the expression of BDNF and TrkB.%目的 观察舒郁宁心汤对慢性应激抑郁模型大鼠海马区脑源性神经营养因子(BDNF)和酪氨酸激酶B(TrkB)表达的影响,探讨其抗抑郁机理.方法 60只SD大鼠随机分为空白组、模型组、氟西汀组及中药(舒郁宁心汤)高、中、低剂量组.复制孤养加慢性轻度不可预见性的应激抑郁模型,造模成功后分组进行药物干预.21 d后采用免疫组化方法观察大鼠海马区BDNF和TrkB表达的变化.结果 与空白组比较,模型组大鼠BDNF和TrkB阳性表达降低(P<0.01);与模型组相比,中药高、中剂量组及氟西汀组大鼠海马区BDNF和TrkB阳性表达提高(P<0.05).结论 舒郁宁心汤可能通过提高海马组织BDNF和TrkB的表达起到治疗抑郁症的作用.
