Objective To establish an HPLC-ELSD method for determination and compare the content of clerosterol 3-O-β-D glucoside in roots and stems of Ilex asprella from different growing areas. Methods HPLC was performed on Kromasil C18 (4.6 mm×250 mm, 5 μm) at 25 ℃, the mobile phase consisted of methanol-water (95 : 5) at a flow rate of 1.0 mL/min. The operating conditions of the ELSD were a nebulizer nitrogen with flow-rate of 1 L/min and an evaporator tube with temperature of 90 ℃. Results The calibration curves of clerosterol 3-O-β-D glucoside was in good linearity in the range of 0.250- 3.992 μg (r=0.9995), the average recovery was 98.2% (n=6). The content of clerosterol 3-O-β-D glucoside in the roots was in the range of 0.114-0.303 mg/g and was in the range of 0.061-0.224 mg/g in the stems of Ilex asprella from different growing areas. Conclusion The method is simple, quick and accurate, it can be used for the quality control of Ilex asprella. The content level of clerosterol 3-O-β-D glucoside in the root is higher than that in the stem of Ilex asprella.%目的 建立岗梅根、茎中赪酮甾醇3-O-β-D 葡萄糖苷的含量测定方法,比较不同产地岗梅根、茎中赪酮甾醇3-O-β-D 葡萄糖苷的含量.方法 采用高效液相色谱法.色谱柱为Kromasil C18 柱(4.6 mm×250 mm,5 μm),流动相为甲醇-水(95∶5),流速为1.0 mL/min,柱温25 ℃.蒸发光散射检测器参数:漂移管温度为90 ℃,氮气流速为1 L/min.结果 赪酮甾醇3-O-β-D 葡萄糖苷在0.250~3.992 μg 范围内呈良好的线性关系(r =0.999 5),平均回收率为98.2%,RSD=2.0%(n =6).岗梅根中含量在0.114~0.303 mg/g 之间,岗梅茎中含量在0.061~0.224 mg/g 之间.结论 本方法简单快捷、准确可靠,可用于岗梅药材的质量控制.岗梅根中赪酮甾醇3-O-β-D 葡萄糖苷含量比岗梅茎高.
