目的 探讨E-cadherin、β-catenin、肌球蛋白轻链激酶(myosin light chain kinase,MLCK)蛋白在高甘油三酯血症相关性急性胰腺炎(hypertriglyceridemia associated acute pancreatitis, HTGP)早期大鼠模型中的表达及意义.方法 144只雄性SD大鼠随机分为普通饲料组(48只),高脂饲料组(96只),喂养4周,普通饲料组随机分为2个亚组:N组、ANP组,高脂饲料组随机分为4个亚组:HTG组、HTGP组、HTG+ML-7组、HTGP+ML-7组. N组、HTG组、HTG+ML-7组为假手术组,ANP组、HTGP组、HTGP+ML-7组通过十二指肠乳头逆行胆胰管注射质量浓度为40 g/L的牛黄胆酸钠建立急性坏死性胰腺炎模型,并于造模后3、6、12 h处死大鼠,应用HE染色观察胰腺形态学的变化,检测血清淀粉酶含量,免疫组织化学技术检测胰腺组织E-cadherin、β-catenin及MLCK蛋白的定位及表达.结果 高脂饲料组与普通饲料组相比,TG水平含量显著升高(P0. 05);同时点造模组血清Ca2+比较,差异无统计学意义(P>0. 05);HTGP 组与 ANP 组各时点相比,E-cadherin、β-catenin、MLCK 蛋白含量显著升高(P0. 05). Expressions of E-cadherin, β-catenin, MLCK in HTGP group showed statistically significant differences compared with ANP group (P<0. 05). Expressions of E-cadherin, β-catenin and MLCK were positively correlated with pancreatic pathological scores (r1=0. 644, r2=0. 474, r3=0. 685, P<0. 05). The degree of pancreatic injury in HTGP+ML-7 group treated with MLCK inhibitor ML-7 was lower than that in HTGP group ( P<0. 05), and the expressions of E-cadherin, β-catenin and MLCK proteins were decreased (P<0. 05). Conclusion The expressions of E-cadherin, β-catenin and MLCK are increased in the early of acute necrotizing pancreatitis, and the same point HTGP is higher than that in ANP group. MLCK may change the adhesion junction and participate in the formation of HTGP.
