Smad3和Smad7在梗阻性黄疸大鼠肝纤维化形成过程中的表达及意义

             

摘要

目的:研究梗阻性黄疸大鼠肝纤维化形成进程中Smad3与Smad7表达的变化及其对肝纤维化的意义.方法:Wistar大鼠60只,随机分成模型组和对照组.于术后第1天及术后7d、14d、28 d及35d5个时间点模型组和对照组处死大鼠各6只.进行肝组织病理学观察,免疫组织化学法测定肝组织中Smad3、Smad7的表达情况并行半定墨分析.结果:与对照组相比,模型组大鼠7、14、28、35 d肝纤维化程度逐渐增强,Smad3明显升高并有统计学意义(P<0.001),Smad7明显降低并有统计学意义(P<0001);而且模型组5个时间点比较均有统计学意义(P<0.001).同时,Smad3与Smad7呈负的直线相关(r=-0.951,P<001).结论:随着胆道梗阻时间延长,肝纤维化程度逐渐加重,肝组织中Smad3表达水平显著增加,Smad7表达水平下降,Smad3与Smad7呈负的直线相关.抑制Smad3或增强Smad7的表达可能是防治肝纤维化的新途径.%Objectives To study the expression of Smad3 and Smad7 in hepatic fibrosis of obstructive jaundice in rats and its significance in hepatic fibrosis investigation. Methods: 60 Wistar rats were randomly divided into two groups: model group and control group, the model group was induced by bile commen duct ligation. In the Id, 14& 28d, 35d of the experiment, rat liver tissues were taken for morphological analysis and the stage of hepatic fibrosis was observed under light microscope. The protein expressions of Smad3 and Smad7 in liver tissue were determined with immunohistochemistey assay. Results* Morphological analysis showed that the function of liver worsed and the liver fibrosis was gradually increased in model group, the expression of Smad3 was increased in liver, compared with those in control group (P<0.001), While the expression of Smad7 was decreased in liver, compared with those in control group (P<0.001), There were statistical significant among 1 d,7d, 14d,28d,35d. Meanwhile, there was negative linear correlation between Smad3 and Smad 7 (r=-0.951,P<0.001). Conclusion: During hepatic fibrosis induced by bile commen duct ligation in rats, the expression of Smad3 is increased ,while Smad7 is decreased, there is negative linear correlation between Smad3 and Smad 7. Smad3 and Smad 7 have a close relationship with heptic fibrosis.
