脂肪肉瘤中AFAP1L1和AFAP1的表达及意义

摘要

目的 探讨AFAP1 L1和AFAP1在脂肪肉瘤(liposarcoma)中的表达及临床意义.方法 采用免疫组化MaxVision法分别检测62例脂肪肉瘤和20例良性脂肪瘤中AFAP1L1和AFAP1的蛋白表达；采用原位杂交法分别检测62例脂肪肉瘤中AFAP1L1和AFAP1的mRNA表达.结果 AFAP1L1和AFAP1的蛋白在脂肪肉瘤中的阳性率分别为71.0％ (44/62)和40.3％ (25/62),高于在良性脂肪瘤中的表达(P＜0.05)；AFAP1L1和AFAP1的mRNA在脂肪肉瘤中阳性率分别为69.4％(43/62)和29.0％ (18/62),高于在良性脂肪瘤中的表达(P＜0.05).AFAP1L1和AFAP1的蛋白水平表达与其各自的mRNA表达水平之间具有较好的一致性.脂肪肉瘤中AFAP1L1与AFAP1的表达呈负相关(r=-0.343,P＜0.05).AFAP1L1的表达与肿瘤复发或转移有关(P＜0.01),而与肿瘤的发生部位及不同分化程度无关(P＞0.05).AFAP1蛋白表达与各项临床参数均无关(P＞0.05).结论 AFAP1L1与AFAP1的表达可能与脂肪肉瘤的发生、发展有关,其中AFAP1L1与肿瘤复发或转移关系密切,有望成为判断预后参考指标之一,并可能成为肿瘤基因治疗的新靶点.%Purpose To investigate the expression of AFAP1L1 and AFAP1 and their clinical significance in liposarcoma (LPS). Methods Immunohistochemical method was adopted to detect the expression of AFAP1 LI and AFAP1 proteins in 62 cases of LPS and 20 cases of benign lipoma. In situ hybridization was adopted to detect the expression of AFAP1 LI and AFAP1 on the mRNA level in 62 cases of LPS. Results The positive expression of AFAP1L1 and AFAP1 was 71. 0% (44/62) and 40. 3% (25/62) , respectively, which was higher than that of benign lipoma (P 0. 05 ) and differentiation degree (P > 0. 05). No relationship was found between AFAP1 positive expression and clinicopathological features ( P > 0. 05 ) . Conclusion The expression of AFAP1 LI and AFAP1 is significantly involved in the carcinogenesis, progression of LPS, and AFAP1 LI is closely related to the invasion or metastasis of LPS and hopeful to become one of the diagnosis and prognostic indicators of the tumor. It will also provide a new target for the gene therapy of the tumor.
