地黄饮子加减方对MCAO模型大鼠HPA轴影响的实验研究

     

摘要

Objectives:Through measuring the level of HPA axis in plasma of MCAO rat models at different time points and after the stomach irrigation of Dihuang YinZi,we observe the variation characteristics of the level and the intervention effects of Dihuang YinZi. Methods:By using RIA (radioimmunoassay), we measured the level of HPA axis in plasma of MCAO rat models 6 and 48 hours after cerebral ischemia, and 7 days after perfusing with Dihuang YinZi into rats'stomach.Results:①In 6 hours after cerebral ischemia, the level of HPA axis in model group was lower than that of normal group,and the function of HPA axis was in inhibitory state. ②48 hours after cerebral ischemia, compared with Normal group, the content of ACTH and COR of model group increased significantly and the content of CRH reduced ulteriorly ③ Dihuang YinZi can improve the content of CRH and ACTH in the plasma of MCAO rat models, while reducing content of COl.Conclusion: Modified Dihuang Yinzi can improve content of CRH and ACTH in the plasma of MCAO rat models, while reduce content of COR; its protecting action in acute stage of cerebral ischemia is attributed to improving the function of HPA axis.%目的:通过对MCAO模型大鼠缺血不同时点及地黄饮子灌胃后HPA轴水平的检测,观察其变化特点及地黄饮子的干预效应.方法:采用放射免疫分析法测定MCAO模型大鼠缺血后6h,48h和灌胃地黄饮子7d后的血浆HPA轴水平.结果:①脑缺血6h时模型组HPA轴水平低于正常组,其功能呈抑制状态;②脑缺血48h时,模型组ACTH,COR的含量较正常组明显升高,CRH含量进一步降低;③地黄饮子能够提高MCAO模型大鼠CRH,ACTH的血浆含量并降低COR的含量.结论:地黄饮子加减方能够提高MCAO模型大鼠CRH、ACTH的血浆含量并降低COR的含量,通过改善HPA轴功能的紊乱在脑缺血急性期发挥保护作用.
