五倍子水提取物对菌斑生物膜早期形成过程中结构及细菌活性的影响

         

摘要

目的:应用人工口腔观察五倍子水提取物对菌斑生物膜早期形成过程中结构及细菌活性的影响.方法:选用与致龋相关的三种细菌在人工口腔中形成菌斑生物膜,实验组和对照组分别用2mg/ml五倍子水提取物和生理盐水处理;通过在激光共聚焦显微镜下,特异性荧光染料技术标记死菌和活菌,观察五倍子水提取物对菌斑生物膜早期形成过程中结构和细菌活性的影响.结果:实验组菌斑生物膜厚度降低,细菌呈散在分布,且死菌面积增大,活菌面积减小,细菌活性明显低于对照组.结论:五倍子水提取物能杀灭菌斑生物膜内的细菌,降低生物膜的活性,改变其结构的完整性.%Objective To observe the effection of gallnut water extract on the morphology and bacterial activity of bacteria biofilm during the early-forming period. Methods In an artificial mouth, three cariogenic bacteria were used to form plaque biofilms. Plaque biofilms were treated with gallnut water extract at a concentration of 2 mg/ml in experimental group,and saline in control group. Bacterias were marked by specific fluorescent dye technique,and observed the structure and bacterial activity of biofilms by confocal laser scanning microscopy. Results Biofilms in experiment group showed decreasing thickness,diffused distribution and less activity. The proportion of dead bacteria increased,and viable bacteria decreased. Conclusion The gallnut water extract can kill bacteria in biofilms,reduce its activity and change the completeness of its structure.
