目的：研究黄芩素对人肺癌细胞A549的增殖抑制作用及细胞周期的影响，并探讨其作用机制。方法采用四甲基偶氮唑盐( MTT )法检测黄芩素对A549细胞生长的抑制作用，采用流式细胞术测定细胞周期变化及细胞凋亡率，并分析EphA2，Survivin及Caspase-3蛋白表达的变化。结果黄芩素可显著抑制A549细胞的生长，且呈时间-剂量依赖关系；黄芩素可阻滞细胞于S期，诱导细胞凋亡率的升高；黄芩素可显著抑制EphA2，Survivin蛋白的表达，同时上调Caspase-3蛋白的表达。结论黄芩素可抑制人肺癌细胞A549的生长，阻滞细胞周期进程，诱导细胞凋亡，其机制可能与下调EphA2和Survivin的表达及促进Caspase-3的表达有关。%Objective To study the inhibiting effects of baicalein on proliferation of human lung carcinoma cell line A549 and its influ-ence on cell cycle,and to explore its molecular mechanism. Methods The MTT assay was used to determine the inhibiting effect of baicalein on the proliferation of A549 cells. The changes of cell cycle and cellular apoptotic rate were detected by the flow cytome-try. The change of expressions of EphA2,Survivin and Caspase-3 protein were analyzed. Results Baicalein could significantly inhibit the growth of A549 cells in time- and dose- dependent manners;baicalein blocked the cells at S phase and induced the increase of apoptotic rate;baicalein significantly down-regulated the expressions of EphA2 and Survivin protein,while up-regulated the expression of Caspase-3 protein in A549 cells. Conclusion Baicalein can inhibit the growth of A549 cells,block the cell cycle progress and in-duce the apoptosis of lung carcinoma cells. Its mechanism may be related with down-regulating the expression of EphA2 and Surviving protein and simultaneously up-regulating the expression of Caspase-3.
