酶放大免疫测定技术法检测丙戊酸药物浓度的动态飘逸校正系数

         

摘要

目的 探讨酶放大免疫测定技术法检测丙戊酸药物浓度的动态飘逸校正系数.方法 以50μg/mL丙戊酸标准水溶液为质控样品,采用德国西门子全自动生化仪,分别在1个试剂盒的第48,108次测试时做质控检测,进行质控检测浓度动态飘逸指数函数的推拟.结果 动态飘逸系数k=(1+0.00066×n0.55535)n,动态飘逸校正系数k′=1/[(1+0.00066×n0.55535)n],并以第1,20,31,97,120,135次质控样品检测浓度验证计算,相对误差在±6%以内.结论 推拟得到的动态飘逸校正系数可用于丙戊酸质控检测浓度的动态校正,有利于丙戊酸血药浓度的准确检测,可减少丙戊酸检测系统不定期定标或质控产生的试剂浪费.%Objective To investigate the dynamic drift correction coefficient for detection of valproic acid concentration by enzyme multi-plied immunoassay technique ( EMIT ) . Methods 50 μg/mL valproic acid standard water solution was taken as the quality control sample. The automatic biochemical analyzer made in Germany Siemens was used to do the quality control test at the 48th,108th tests of a kit, in order to speculate the dynamic drift exponential function of the quality control detection concentration. Results The dynamic drift coefficient was obtained: k=( 1+0. 00066 × n0. 55535 ) n, and the dynamic correction coefficient was obtained: k′=1/ [ ( 1+0. 00066 × n0. 55535 ) n ] . The detection concentration of the 1st, 20th, 31st, 97th 120th and 135th quality control samples were used to verify the calculation, and the relative errors were between -6% and 6%. Conclusion The obtained dynamic drift correction coefficient can be used for the dynamic correction of quality control concentration of valproic acid, which is conducive to the accurate detection of blood concentration of valproic acid, and it can reduce the waste of reagents produced by the irregular calibration or quality control of the valproic acid detection system.
