Objective: To observe the changes of mitochondrial DNA ATPase 6, 8 genes of small intestine epithelial cells in ischemia-reperfusion rats. Methods: 6 rats were randomly divided into the control and ischemia-reperfusion group. Their mtDNA were distilled and the sequences were examined after polymerase chain reaction conducted by using special primer in order to detect mutational sites. Results: The polymorphism existed at some sites of ATPase 6, 8 genes of Wistar rats in China. There was a A7 797 deletion and several sites mutation of ATPase 6,8 genes in ischemia-reperfusion group, namely, A7 779T, T8 515G, T8 520G, C8 535G. Conclusion: The damage of ATPase 6, 8 genes took place after ischemia-reperfusion and this may result in the changes of their coding amino acids.%目的观察缺血再灌注大鼠肠上皮细胞mtDNA ATPase 6,8亚基基因的改变情况.方法将6只Wistar大鼠分成对照组、缺血再灌注组.分别提取mtDNA,采用PCR技术扩增后测序,检测突变点.结果 Wistar大鼠mtDNA ATPase 6,8亚基基因存在多态性,在缺血再灌注组存在a7797缺失和一定数量点突变:A7 779T,T8 515G,T8 520G,C8 535G.结论缺血再灌注后,肠上皮细胞mtDNA ATPase 6,8亚基基因存在损伤,由此可能导致所编码的氨基酸改变.
