牛血清白蛋白-5-磺基水杨酸体系的荧光共振能量转移研究

     

摘要

Fluorescence resonance energy transfer of bovine serum albumin (BSA)-5-sulfosalicyclic acid (SSA)system was studied by fluorescence emission and UV absorption spectra.Results showed that,the fluo-rescence emission spectrum of BSA was quenched by addition of SSA solution and the fluorescence emission peak of BSA emerged a blue shift,but the peak shape of BSA fluorescence spectrum was unchanged.The maxi-mum UV absorption peak of BSA emerged a red shift and its absorbance intensity increased obviously with the increasing of SSA concentration.According to Förster theory of non-radiation energy transfer,the distance be-tween the donor(BSA)and the acceptor(SSA)in BSA-SSA system was calculated as 2.42 nm.%采用荧光发射光谱和紫外吸收光谱研究了牛血清白蛋白(BSA)-5-磺基水杨酸(SSA)体系的荧光共振能量转移。结果表明,SSA 可以猝灭 BSA 的荧光且使 BSA 荧光发射峰蓝移,但峰形未改变;随 SSA 浓度的增大,BSA 紫外吸收光谱的最大吸收峰红移且强度逐渐增强;根据 Förster 非辐射能量转移原理计算得到 BSA-SSA 体系中供体(BSA)与受体分子(SSA)间距离为2.42 nm。
