以羧甲基-β-环糊精( CM-β-CD)作为手性流动相添加剂,于C18柱上建立了高效液相色谱手性流动相添加剂法拆分盐酸美西律对映体的方法。系统的考察了流动相中的添加剂的浓度、有机修饰剂、pH值以及流速对拆分的影响。色谱分离条件:流动相为甲醇、乙腈、水,体积比为15∶70∶15,其中CM-β-CD的浓度为0.5 g/L,pH值为4.28(用三乙胺和冰乙酸调节),流速为0.3 mL/min,检测波长为212 nm,盐酸美西律对映体最佳分离。%Separation of Mexiletine hydrochloride enantiomers by HPLC was established on C18 column using the carboxymethyl-β-cyclodextrin ( CM-β-CD in short) as chiral mobile phase additive. The influences of concentration of chiral mobile phase additive, the organic modifier, the pH value and the flow rate on the enantiomers separation were investigated systematically. The better separation was achieved when the mobile phase consist of methanol-acetoni-trile-water 15∶70∶15(V/V/V), pH=4. 28( adjusted by triethylamine-glacial acetic acid ), containing 0. 5 g· L-1 CM-β-CD, at the flow rate of 0. 3 mL·min-1 . The detection wavelength was set at 212 nm.
