目的 观察钙激活性中电导钾离子通道( IKCa1)在宫颈癌组织中的表达变化及其在宫颈癌HeLa细胞凋亡中的作用.方法 用RT-PCR检测18例正常宫颈组织及15例宫颈癌组织中IKCa1 mRNA的表达;用IKCa1通道的阻断剂克霉唑处理宫颈癌HeLa细胞,用RT-PCR检测细胞IKCa1 mRNA的表达变化,MTT法观察细胞增殖抑制的变化.结果 宫颈癌组织中IKCa1 mRNA的阳性表达率(73.33%)及表达水平(1.14±0.45)明显高于正常宫颈组织(11.11%和0.86±0.23)(P<0.05).克霉唑以浓度和时间依赖的方式抑制HeLa细胞的增殖,下调IKCa1 mRNA的表达水平,诱导细胞凋亡.结论 IKCa1的异常表达可能与宫颈癌相关,降低其表达可能通过诱导细胞凋亡而抑制宫颈癌细胞的增殖.%Objective To investigate the expression of Intermediate-conductance-Ca2+ -activated K+ channels (IKCa1) in cervical cancer and its role in cell apoptosis of cervical carcinoma HeLa cell line. Methods RT-PCR was used to test the expression of IKCal channels in 18 normal cervix specimens, 15 cervical cancer specimens and cervical carcinoma HeLa cells. HeLa cell was exposed to the known inhibitor of IKCal channel clotrimazole. MTT was used to measure the proliferative inhibition of HeLa cell when treated with clotrimazole. Apoptosis was detected by flow cytometer. Results The expression rate and level of IKCal mRNA in cervical carcinoma (73. 33% , 1. 14 ± 0. 45 ) were higher than in normal cervix (11.11%, 0. 86 ±0. 23, P<0. 05). Clotrimazole, caused a both dose-and time-dependent inhibition of cell growth,the IKCal mRNA expression and apoptosis induction in HeLa cells. Moreover, the cell apoptosis was observed. Conclusions The expression of IKCal channels may be closely related to the proliferation of cervical carcinoma, and suppress its expression may inhibit the development of cervical carcinoma through cell apoptosis induction.
