棘豆素对人雄激素非依赖性前列腺癌 PC-3细胞的作用研究

     

摘要

Objective To explore the effect of 2′,4′-dihydroxychalcone (TFC)on androgen-independent prostate cancer PC-3 cells and its possible anti-cancer mechanism.Methods The CCK-8 assay was employed for cellular proliferation measurement,Hoechst 33258 dye and fluorescent microscope for morphological observation,flow cytometry (FCM)for detection of apoptosis and cell cycles, Western blot for analyzing the expression of P27kip1 and PTEN proteins in PC-3 cells treated with TFC of different concentration.Re-sults TFC significantly inhibited the proliferation of PC-3 cells,induced apoptotic morphological features,increased the percentage of apoptosis,arrested cell cycle in G0 /G1 phase,up-regulated the expression of P27kip1 and PTEN proteins in a concentration dependent manner.Conclusions TFC could inhibit the proliferation and induce the apoptosis in PC-3 cells by up-regulating P27kip1 and PTEN protein and arresting cell cycle in G0 /G1 phase.%目的:研究棘豆素(2′,4′-dihydroxychalcone,TFC)对人雄激素非依赖性前列腺癌 PC-3细胞的作用,并探讨其作用机制。方法CCK-8法检测药物对细胞增殖的影响;Hoechst 33258染色、荧光显微镜观察细胞的形态学变化;流式细胞术分析细胞凋亡率和细胞周期的变化;Western blot 检测药物对肿瘤细胞中 P27kip1和 PTEN 蛋白表达的影响。结果TFC 能显著抑制PC-3细胞的增殖,细胞呈现凋亡形态学改变,凋亡率明显增加,细胞周期阻滞于 G0/G1期。此外,P27kip1和 PTEN 蛋白表达量明显增加。结论TFC 通过诱导 PC-3细胞凋亡抑制肿瘤细胞增殖,可能与其 G0/G1细胞周期阻滞,以及 PTEN 和 P27Kip1蛋白表达上调有关。
