IL-17与 TNF-α对 HaCaT 细胞 IL-17R、p-p38 MAPK和炎症因子表达的影响

摘要

Objective To investigate the co-effect of IL-17 and TNF-αon the expressions of IL-17R,p-p38 MAPK in skin keratinocyte cell line HaCaT cells,and on the levels of IL-6,IL-8,MIP-3αsecreted by HaCaT cells.Methods RT-PCR method was applied to ana-lyze the expression of IL-17R mRNA in HaCaT cells treated with IL-17 and TNF-αin combination;Western blot method to study the expression of p-p38 MAPK;ELISA assay to analyze the level of inflammatory factors IL-6,IL-8 and MIP-3α.Results Co-effect of IL-17 and TNF-αcould increase the expressions of IL-17R and p-p38 MAPK in HaCaT cells,as well as increase the levels of IL-6,IL-8, MIP-3αsecreted by HaCaT cells.Conclusions Co-effect of IL-17 and TNF-αcan promote the expression of IL-17R,stimulate the phosphorylation of p38 MAPK,and increase the expressions of IL-6,IL-8,MIP-3α.So co-effect of IL-17 and TNF-αhave played a sig-nificant proinflammatory role.%目的：考察 IL-17与 TNF-α共作用对皮肤角质形成细胞系 HaCaT 细胞 IL-17R、p-p38 MAPK 表达的影响，及其对 HaCaT细胞分泌 IL-6、IL-8、MIP-3α等炎症因子的影响。方法采用 RT-PCR 方法检测 IL-17与 TNF-α共作用对皮肤角质形成细胞IL-17R mRNA 表达的影响；采用 western blot 方法检测 IL-17与 TNF-α共作用对 HaCaT 细胞表达 p-p38 MAPK 的影响；采用ELISA 法检测 IL-17与 TNF-a 共作用对 HaCaT 细胞分泌炎症因子 IL-6、IL-8及 MIP-3α的影响。结果IL-17与 TNF-α共作用可上调 HaCaT 细胞表达 IL-17R 和 p-p38 MAPK，以及上调 HaCaT 细胞 IL-6、IL-8、MIP-3α等炎症因子的分泌。结论IL-17与TNF-α共作用可通过上调皮肤角质形成细胞 IL-17R 的表达，促进 p38 MAPK 的磷酸化过程而具有明显致炎作用。
