红树莓根茎水煎剂对昆明小鼠的急性毒性及安全性实验研究

         

摘要

Objective:To study the acute toxicity and safety of raspberry rhizome decoction on Kunming mice. Methods:LD50 test of raspberry rhizome decoction was carried out at the dose of 222. 4 g/(kg·d). The general state, body weight and death of mice were observed after being given the maximum dose of 444. 8 g/(kg·d). The blood biochemical inde-xes and main viscera indexes were detected, and the pathological changes of main organs were observed. Results: It showed that the LD50 test could not be measured. The maximum dosage test showed that no death or toxicity happened in the mice, and the general condition of the mice was good. Body weight of the mice was significantly reduced only on the first day after the administration of raspberry rhizome decoction, compared to the control group(P<0. 01); but there was no significant difference between the experimental group and the control group in terms of weight gain on the fourth and seventh day(P>0. 05). There was also no significant difference between the experimental group and the control group in terms of blood biochemistry test and main organ index ( P>0 . 05 ) . The pathological examination showed no morphological changes of main organs after the administration of raspberry rhizome decoction. Conclusion: Raspberry rhizome decoction has no obvious acute toxicity to Kunming mice, which has high safety.%目的:研究红树莓根茎水煎剂对昆明小鼠的急性毒性及安全性.方法:预实验,即半数致死量实验,给小鼠灌胃药量为222.4 g/(kg·d),测量红树莓根茎水煎剂的半数致死量.最大给药量实验,给小鼠灌胃药量为444.8 g/(kg·d),观察小鼠的一般状态、体质量和死亡等情况;检测小鼠血液中的生物化学指标和主要脏器指数;观察主要脏器的病理形态学变化.结果:预实验结果显示无法测出红树莓根茎水煎剂的半数致死量.最大给药量实验结果显示:实验小鼠无中毒死亡情况、一般状态良好,体质量仅在灌胃后第1天与对照组比较明显减轻(P<0.01),第4,7天体质量增加与对照组比较无显著差异(P>0.05);实验小鼠血液生物化学检查结果、主要脏器指数检查结果与对照组比较,均无统计学差异(P>0.05);病理形态学检查未发现红树莓根茎对小鼠主要脏器产生形态结构的异常改变.结论:红树莓根茎水煎剂无毒,对昆明小鼠无明显的急性毒性作用,具有较高的安全性.
